摘  要:目的通过研究正常大鼠以及双侧颈总动脉结扎后大鼠脑血流量的变化,探讨疲劳运动对脑卒中大鼠脑血流及学习记忆的影响。方法 120只雄性Wistar大鼠根据体重随机分为对照组(SO),过度运动组(OE),双侧颈总动脉结扎组(2-VO),双侧颈总动脉结扎并过度运动组(2-VO+OE)。采用大鼠跑台进行疲劳运动训练,采用双通道激光多普勒脑血流仪对大鼠的额叶皮质脑血流量进行测定,Morris水迷宫实验对大鼠的学习记忆情况进行测定。结果 与SO组相比,OE组脑血流量显著升高(P<0.05),但2-VO组及2-VO+OE组脑血流量均下降(P<0.01),与2-VO组比较2-VO+OE组脑血流量显著下降(P<0.01);与SO组相比,2-VO+OE组红细胞运动速度下降(P<0.01),其余各组并无明显变化(P>0.05);红细胞聚集度4组之间并无明显变化(P>0.05);回光强度4组之间并无明显变化(P>0.05);在水迷宫实验中,与SO组比较,2-VO组与2-VO+OE组逃避潜伏期显著增加(P<0.01),穿台次数显著减少(P<0.01),其中2-VO+OE组潜伏期增加的更为明显,但与2-VO组比较无显著性差异(P>0.05),平均速度各组之间并无明显变化(P>0.05)。结论 脑缺血后大鼠脑血流量会显著下降,造成学习记忆障碍。缺血前的疲劳运动会增大缺血后大鼠脑血流量的下降幅度,且会导致学习记忆障碍的加重。Objective To investigate the effect of fatigue exercise on cerebral blood flow and learning and memory in stroke rats by studying the changes of cerebral blood flow in normal rats and bilateral common carotid artery ligation. Methods One hundred and twenty male Wistar rats were randomly divided into control group(SO),excessive exercise group(OE),common carotid artery ligation group (2-VO),bilateral common carotid artery ligation and excessive exercise group according to body weight( OE+2-VO). Excessive exercise was performed on a rat running platform for fatigue exercise training. Cerebral blood flow in rats was measured using a dual-channel laser Doppler cerebral blood flow meter. Morris water maze test was used to determine the learning and memory status of rats. Results Cerebral blood flow was significantly higher in the OE group than in the SO group( P < 0.05 ),but cerebral blood flow was lower in the 2-VO group and the 2-VO+OE group ( P < 0.01 ),compared with 2-VO group 2-The cerebral blood flow in the VO+OE group was significantly decreased ( P < 0.01 );compared with SO,the red blood cell movement speed in the 2-VO+OE group was decreased( P < 0.01 ),and there was no significant change in the other groups( P > 0.05 );There was no significant change between the two groups( P > 0.05 ). There was no significant change between the four groups( P > 0.05 ). In the water maze experiment,compared with the SO group,2-VO group and 2-VO+OE group escaped. The latency period increased significantly ( P < 0.01 ),and the number of punctures decreased significantly( P < 0.01 ). The latency of 2-VO+OE group increased more significantly,but there was no significant difference compared with 2-VO group ( P > 0.05 ). There was no significant change in the average speed between groups( P > 0.05 ). Conclusion Cerebral blood flow will be significantly decreased after stroke in rats. Pre-stroke fatigue exercise will increase the decrease of cerebral blood flow after stroke,and it will lead to increased learning and memory impairment
