摘　　要：目的评价VEGF-A、C、D在胃癌腹水瘤模型中疗效预测价值。方法NUGC-4-luc细胞腹腔接种28只雌性裸鼠,1周后随机分组:Group1:顺铂(1mg/kg)d1~3+恩度(8mg/kg)d4~7;Group2:恩度(8mg/kg)d1~4+顺铂(1mg/kg)d5~7;Group3:恩度(8mg/kg)+顺铂(1mg/kg)d1,4,7;Group40.9%氯化钠溶液50μld1-7,给药2周停药1周后处死。ELISA检测积液上清VEGF-A、C、D蛋白含量;腹膜结节包被蜡块后,IHC方法检测VEGF-A、C、D表达。结果4组腹水量(ml):Group4>Group1>Group2>Group3。4组腹膜结节数量:Group4>Group2>Group3>Group1。腹水上清VEGF-A在Group4>Group1>Group2>Group3。腹膜结节VEGF-A,C,D均在癌细胞胞浆着色,VEGF-A表达Group4>Group2>Group1,差异有统计学意义(P<0.05)。VEGF-A含量与腹水量高度相关(r=0.994,P<0.01)。VEGF-A表达与腹膜结节数量高度相关(r=0.972,P<0.01)。结论联合治疗组对腹水及腹膜结节VEGF-A抑制作用均最强,恩度序贯顺铂组对腹水VEGF-A抑制作用强,顺铂序贯恩度组对腹膜结节VEGF-A抑制作用强。VEGF-A不仅预测腹水量而且预测腹膜结节的变化,提示其可作为整体治疗疗效的潜在指标。Objective To evaluate the predivtive value of VEGF-A,C,D for the therapeutic effects of endostar in different medication ways on gastric cancer ascites models in nude mice.Methods The NUGC-4 gastric cancer cells transfected with luciferase gene(NUGC-4-Luc)were injected peritoneally into 28 nude mice.One week later,the mice were randomly group 1:cisplatin(1mg/kg)d1~3+endostar(8mg/kg)d4~7;group 2:,endostar(8mg/kg)d1~4+cisplatin(1mg/kg)d5~7;group 3:endostar)8mg/kg)+cisplatin(1mg/kg)d1,4 and 7;group 4:0.9%sodium chloride solution 50μl,d1~7.The mice were sacrificed after 2-week medication,the levels of VEGF-A,C,D in supernatant fluid of ascites were detected by ELISA,and the levels of VEGF-A,C,D in peritoneal nodules were detected by IHC.Results The comparison of volume of ascites in four groups(ml):group 4(4.87±0.45)>group1(3.1±0.53)>group2(2.0±0.08)>group3(1.8±0.16).The number of peritoneal nodules:group 4(33.75±2.5)>group 2(21.66±5.77)>group 3(18.75±2.5)>group 1(8.75±4.78).The levels of VEGF-A in supernatant showed that group4>group1>group2>group3(P<0.05).The VEGF-A,C,D proteins in peritoneal nodules were stained in cancer cell cytoplasm,and the levels of VEGF-A were group4>group2>group1(P<0.05).Moreover the VEGF-A protein content was closely correlated with the volume of ascites(r=0.994,P<0.01),and the VEGF-A protein expression was closely correlated with the number of peritoneal nodules(r=0.994,P<0.01).Conclusion The inhibitory action of combination treatment on VEGF-A expression in ascites and peritoneal nodules of nude mice models is the strongest,and the inhibitory action of endostar sequential cisplatin on VEGF-A expression in ascites of nude mice models is the strongest,and the inhibitory action of cisplatin sequential endostar on VEGF-A expression in peritoneal nodules of nude mice models is the strongest,which can be used as potential indexes to evaluate the whole therapeutic effects.
