摘  要:胃泌素释放肽受体(Gastrin-releasing peptide receptor,GRPR)属于G蛋白偶联受体家族,主要分布于中枢神经系统、脊髓和胃肠道组织,通过与其配体胃泌素释放肽(Gastrin-releasing peptide,GRP)结合发挥生物学作用。GRPR参与人体多项生理功能调节过程,在体内具有重要生理学功能。研究发现,GRPR在多种肿瘤中呈现高表达,如多形性胶质母细胞瘤、肺癌、乳腺癌、前列腺癌、胰腺癌等,并参与了这些肿瘤的发生和发展。同时大量实验表明,通过使用GRPR抑制剂、靶向GRPR的反义寡核苷酸疗法,起到抑制肿瘤的目的。因此,GRPR日渐成为肿瘤研究、诊断和治疗的热点。该文阐述了GRPR的特点及生物学功能,综述了GRPR在肿瘤中的研究及靶向GRPR的治疗策略,将为临床肿瘤治疗提供思路。Gastrin-releasing peptide receptor( GRPR) belongs to the G protein coupled receptor family,which is mainly distributed in the central nervous system,spinal cord and gastrointestinal tissues. GRPR binds to its ligand gastrin-releasing peptide( GRP) to exert biological effects. GRPR participates in the regulation of many physiological functions of the human body and has important physiological functions. The studies have found that GRPR is highly expressed in a variety of tumors,such as pleomorphic glioblastoma,lung cancer,breast cancer,prostate cancer,pancreatic cancer,etc.,and is involved in the development and progression of these tumors. A large number of studies have reported that GRPR inhibitors and antisense oligonucleotide therapy can inhibit tumor cell proliferation. Therefore,GRPR is becoming a hot spot for cancer research,diagnosis and treatment. This article describes the characteristics and biological functions of GRPR,reviews the research of GRPR in tumors and the treatment strategy of targeting GRPR,and will provide ideas for its clinical application.
