摘  要:在陆生高等植物叶绿体中,RNA编辑普遍存在.为探究甘薯属植物叶绿体RNA编辑的起源与进化及基因的转录调控,以甘薯属六倍体栽培种徐薯18以及近缘野生种三浅裂野牵牛(I.trifida)和五爪金龙(I.cairica)为材料,采用生物信息学等方法,对5个蛋白编码基因RNA编辑位点进行分析鉴定.结果共预测到分布于3个物种中5个蛋白编码基因的94个编辑位点,均为C到U的碱基替换.通过验证,共27个真实存在的RNA编辑,编辑包括碱基的缺失、插入与替换,氨基酸转变类型增加.通过比较知,甘薯与2个野生种之间RNA编辑位点的保守性较低.另外,仅徐薯18中ndhB基因编辑后编码的蛋白质跨膜结构域增加,但大部分RNA编辑对蛋白质二级结构影响不大.RNA editing is ubiquitous in chloroplast of higher terrestrial plants. To explore the origin and evolution of chloroplast RNA editing sites of Ipomoea (L.) and its gene transcription regulation,Xushu18, I.trifida and I.cairica were used as experimental materials. The RNA editing sites of five protein coding genes were analyzed and identified by bioinformatics prediction and other methods. Results showed that 94 editing sites distributed in 5 protein coding genes in three species were predicted, all of which were base substitution from C to U, and the amino acid transformation forms were diverse. Verification found 27 real RNA edits, including base insertion,deletion and substitution, and the amino acid transformation types increased. By comparison, the conservation of RNA editing sites between sweet potato and two wild species was lower. In addition, only the transmembrane domain of protein encoded by ndhB gene editing in Xushu 18 increased, but most RNA editing had little effect on the secondary structure of protein.
