摘 要:目的探讨天麻超微粉细胞破壁率的测定方法,评价超微粉碎后天麻粉末的细胞破碎程度。方法采用显微制片细胞计数法,以天麻粉末中的薄壁细胞为评价指标,确定天麻粉末质量与薄壁细胞数目的线性关系,通过对比天麻常规粉末与超微粉薄壁细胞的数量,计算天麻超微粉的细胞破壁率。结果天麻粉末中薄壁细胞可清晰观察并计数,薄壁细胞数目与粉末的质量成正相关,在3.8124~22.8744mg范围内线性关系良好,天麻粉末经超微粉碎后细胞已完全破碎,超微粉的细胞破壁率可达99%以上。结论天麻粉末显微制片细胞计数法测定天麻超微粉细胞破壁率具有可行性和科学性,能够为评价超微粉的细胞破碎程度提供参考和依据。Objective To explore the determining method of cell wall-broken rate of Gastrodia elata micropowde,and to evaluate the degree of cell breakage of Gastrodia elata micropowde.Methods The parenchyma cells in Gastrodia elata powder were taken as the evaluation index,the linear relationship between the quality of Gastrodia elata powder and the number of parenchyma cells was determined by microscopic cell counting method.By comparing the number of parenchyma cells of Gastrodia elata conventional powder with that of ultrafine powder,the cell wall-broken rate of ultrafine powder of Gastrodia elata was calculated.Results The parenchyma cells in Gastrodia elata powder could be clearly observed and counted,the number of parenchyma cells was positively correlated with the quality of Gastrodia elata powder,and the linear relationship was good in the range of 3.8124-22.8744 mg.The cells of Gastrodia elata powder have been completely broken after ultrafine comminution,the wall-broken rate of Gastrodia elata micropowder was over 99%.Conclusion It is feasible and scientific to determine the wall-broken rate of Gastrodia elata micropowder by microscopic cell counting method,and provide reference and basis for evaluating the degree of cell breakage of ultrafine powder.
