摘  要:初步筛选了地胆粉的最佳用酶及通过了MTT法测定酶解产物的抗肿瘤活性。模拟体外胃肠道消化系统以胃蛋白酶、弹性蛋白酶、胰蛋白酶、糜蛋白酶4种蛋白酶进行酶解反应,采用MTT法检测酶解产物对肝癌细胞BEL-7402、乳腺癌细胞MCF-7的抑制效果。结果弹性蛋白酶、胃蛋白酶、糜蛋白酶的水解度分别为2.84%、13.11%、10.31%,其中胰蛋白酶的酶解液对肝癌细胞BEL-7402的抑制率明显高于对照组,且成剂量依赖性,其他酶解液的抑制作用不显著。胰蛋白酶的酶解液对乳腺癌细胞MCF-7也有一定的抑制作用,其他酶解液无效果。胰蛋白酶是地胆粉抑制肝癌细胞的最佳酶,酶解度与抑制肿瘤细胞生长没有直接关系。To screen the optimal enzyme of Didan powder and determine the anti-tumor activity of the product by MTT assay, four kinds of proteases including pepsin, elastase, trypsin and chymotrypsin were used to simulate the enzymatic hydrolysis reaction of the gastrointestinal digestive system in vitro. The inhibitory effect of the products of enzymatic hydrolysis on liver cancer cells bel-7402 and breast cancer cells McF-7 was detected by MTT assay. The degrees of hydrolysis of elastase, pepsin and chymotrypsin were 2.84%, 13.11% and 10.31%, respectively. The inhibitory rate of trypsin hydrolysate on liver cancer cell bel-7402 was significantly higher than that of the control group, and the inhibitory effect of other hydrolysates was dose-dependent. The enzymatic hydrolysate of trypsin also had certain inhibitory effect on McF-7 of breast cancer cells, while other enzymatic hydrolysates had no effect.Trypsin was the best enzyme to inhibit hepatocellular carcinoma cells, and the degree of enzymatic hydrolysis was not directly related to the inhibition of tumor cell growth.
