摘  要:目的 探讨晚期氧化蛋白产物(AOPPs)对小肠上皮细胞(IEC)线粒体膜电位(MMP)的影响及其相关机制。方法 体外培养大鼠小肠隐窝上皮细胞系(IEC-6),以体外制备的AOPPs进行处理,采用流式细胞仪检测MMP和活性氧(ROS)的变化,Western blotting检测晚期糖基化终产物受体(RAGE)的表达。结果 AOPPs可显著降低IEC-6细胞的MMP,且具有时间依赖效应;AOPPs明显上调IEC-6细胞RAGE的蛋白表达及细胞内ROS水平,通过RAGE中和抗体和超氧化物歧化酶SOD分别阻断膜受体和ROS信号传递均可逆转AOPPs诱导的IEC-6细胞MMP下降效应。结论 AOPPs通过细胞膜受体RAGE和细胞内ROS介导IEC-6细胞MMP下降,从而引起线粒体功能障碍,使细胞出现功能异常。Objective To investigate the effect and related mechanism of advanced oxidation protein products (AOPPs) on mitochondrial membrane potential (MMP) in intestinal epithelial cells (IEC). Methods Cultured immortalized rat intestinal epithelial (IEC-6) were treated with AOPPs,flow cytometry was used to detect the changes of MMP and reactive oxygen species (ROS),the expression of advanced glycosylation end product receptor (RAGE) was detected by western blotting. Results Compared with the negative control group,the MMP of AOPPs group was significantly dropped in IEC-6 cells. AOPPs could obviously up-regulate the expression of RAGE and the production of ROS. Blocking the membrane receptor and ROS signal transduction by RAGE neutralizing antibody and superoxide dismutase (SOD) respectively,could reverse the AOPPs-induced MMP decrease in IEC-6 cells. Conclusion AOPPs reduce the MMP of IEC-6 cells through RAGE and ROS,which results in mitochondrial dysfunction and abnormal cell function.
