摘 要:目的:研究山慈菇水提取物对人胃癌细胞(未分化)(Human Gastric Cancer Cell Lines,HGC-27)的抑制作用及其机制。方法:用四甲基偶氮唑盐(Methyl Thiazolyl Tetrazolium,MTT)法检测不同剂量的山慈菇水提取物对胃癌HGC-27的增殖抑制作用。免疫印迹法检测各浓度组48 h时PI3K/Akt信号通路中的重要蛋白Akt及其磷酸化形式p-Akt的蛋白水平。结果:48 h的时候,不同浓度的山慈菇水提取物对胃癌HGC-27均有抑制作用,且有一定的浓度依赖性和时间依赖性。随着浓度升高,时间的推移,p-Akt/Akt值呈降低趋势,差异有统计学意义(P<0.05)。结论:山慈菇水提取物对胃癌HGC-27增殖起抑制作用,同时与药物浓度、给药时间有一定的相关性,其可能通过抑制PI3K/Akt通路中的Akt活性实现。Objective: To study the inhibitory efficacy of water extract of Shancigu(山慈菇) on gastric cancer HGC-27 and its mechanism. Methods: MTT assay was used to detect the inhibitory effect of different doses of water extract of Shancigu on the proliferation of gastric cancer HGC-27. Western blotting was used to detect the protein levels of the important protein Akt and its phosphorylated form p-Akt in the PI3 K/Akt signaling pathway at 48 h in each concentration group. Results: At 48 h, different concentrations of Shancigu water extracts inhibited gastric cancer HGC-27 in a concentration and time-dependent manner. As the concentration increased, the p-Akt/Akt value decreased with time, and the difference was statistically significant(P<0.05). Conclusion: The water extract of Shancigu can inhibit the proliferation of gastric cancer HGC-27, and it has a certain dependence on drug concentration and administration time, which may be achieved by inhibiting Akt activity in PI3 K/Akt pathway.
