摘 要:目的:探讨长链非编码 RNA(lncRNA)HOX转录本反义 RNA(HOTAIR)在接受调强放射治疗鼻咽癌中的表达及其临床意义。方法:收集20例鼻咽癌患者肿瘤组织及对应癌旁组织的新鲜标本,279例经病理学证实接受调强放射治疗的鼻咽癌患者的组织标本、临床病理特征及随访资料。应用 RT qPCR法检测鼻咽癌组织中HOTAIR的表达情况,并对临床病理资料和随访资料进行统计分析。结果:20例鼻咽癌及其配对的正常鼻咽黏膜上皮组织中,75%(15/20)鼻咽癌组织 lncRNAHOTAIR的表达水平明显高于正常鼻咽组织(P<0.01);在 279例鼻咽癌患者组织中,有59.9%(167/279)鼻咽癌组织 HOTAIR表达升高;HOTAIR的高表达与 T和 N分期密切相关(P<0.05),但与患者的年龄、性别、病理类型和 M分期没有明显相关性;与 lncRNAHOTAIR低表达组相比,HO TAIR高表达组的鼻咽癌患者疾病无进展生存率和总生存率明显下降(P<0.05)。结论:lncRNAHOTAIR在鼻咽癌中表达水平升高,且与鼻咽癌患者的不良预后明显相关。AIM: To investigate the expression and clinical significance of long noncoding RNA HOX transcript antisense RNA (HOTAIR) in nasopharyngeal carcinoma patients receiving intensity-modulated radiotherapy. METHODS: The clinical data and tissue samples of 279 patients of pathologically confirmed nasopharyngeal carcinoma receiving intensity-modulated radiotherapy were collected. The expression of HOTAIR was detected by RT-qPCR. The relationship between the expression of HOTAIR in nasopharyngeal carcinoma tissues and clinicopathological features of the patients was analyzed using χ2 test. The relationship between the expression of HOTAIR and prognosis of the patients with nasopharyngeal carcinoma was analyzed based on univariate and multivariate analyses. RESULTS: The results of RT-qPCR indicated that the expression of HOTAIR in nasopharyngeal carcinoma tissues was significantly higher than that in adjacent normal tissues ( P <0.01). In 279 cases of nasopharyngeal carcinoma, high expression of HOTAIR was found in 59.9%(167/279) cases. The over-expression of HOTAIR was closely related to tumor size and lymph node metastasis ( P <0.05). Univariate and Cox multivariate analyses showed that high expression of HOTAIR decreased the progression-free survival and overall survival of nasopharyngeal carcinoma patients. CONCLUSION: HOTAIR was highly expressed in nasopharyngeal carcinoma tissues, and it is an independent prognostic factor in nasopharyngeal carcinoma patients.
