摘  要:目的研究Ⅱ类组氨酸去乙酰基酶抑制剂(HDACIs)制滴菌素A(TSA)在抑制角膜移植排斥反应中的作用。方法通过增生抑制试验和Treg细胞诱导实验评价TSA对效应性T细胞的抑制率及诱导调节性T细胞比例,同时观察TSA治疗后小鼠角膜植片存活率。结果体外增生抑制试验中暴露在1 ng/ml、5 ng/ml、10 ng/ml TSA中细胞的增生率分别为(74.60±0.03)%、(26.27±0.01)%、(25.64±0.03)%。体外Treg细胞诱导实验中暴露在5 ng/ml和20 ng/ml TSA中CD25+Foxp3+细胞的比率为(3.19±1.32)%和(7.73±1.52)%。体内实验中TSA治疗组小鼠角膜植片的存活率最高。结论TSA治疗后小鼠角膜移植片存活率提高。Objective To investigate the role of class Ⅱ histidine deacetylase inhibitor (HDACIs) Trichostatin A (TSA) in inhibiting corneal graft rejection. Methods The inhibition rate of TSA on effector T cells and the ratio of TSA-induced regulatory T cells were evaluated with proliferation inhibition assay and Treg cell induction assay, and the survival rate of corneal grafts after TSA treatment was observed. Results The proliferation rates of cells exposed to 1 ng/ml, 5 ng/ml, and 10 ng/ml TSA in the in vitro proliferation inhibition test were (74.60±0.03)%,(26.27±0.01)%, and (25.64±0.03)%, respectively. The ratio of CD25+ Foxp3+ cells exposed to 5 ng/ml and 20 ng/ml TSA in vitro was (3.19±1.32)% and (7.73±1.52)%. In vivo experiment, the survival rate of corneal grafts was the highest in TSA treatment group. Conclusion The survival rate of mouse corneal grafts is improved after TSA treatment.
