摘  要:目的研究乙酰辅酶A羧化酶(Acetyl-CoA carboxylas 1,ACC1)在卵巢癌细胞中的表达及其在细胞增殖调控中的作用。方法用实时荧光定量PCR(quantitative real-timePCR,qRT-PCR)与Western Blot方法,分析10例卵巢癌患者癌与癌旁组织中ACC1表达,以明确卵巢癌细胞中ACC1的表达是否发生了异常改变。小干扰RNA(small interference RNA,siRNA)下调卵巢癌细胞SKOV3中ACC1表达后,分别用MTS与EDU实验分析ACC1对细胞增殖的影响;用流式细胞术分析ACC1对细胞凋亡的影响。结果卵巢癌组织中ACC1的mRNA与蛋白表达水平发生上调(P<0.01);下调ACC1表达可显著抑制卵巢癌细胞的增殖(P<0.01);下调ACC1表达对卵巢癌细胞的凋亡无明显影响(P>0.05)。结论ACC1在卵巢癌组织中表达上调并促进卵巢癌细胞的增殖,提示ACC1可能是潜在的卵巢癌治疗的分子标志物。Objective To evaluate the expression of Acetyl-CoA carboxylas 1(AAC1) and its promotive effect on the proliferation of ovarian cancer cells. Methods Quantitative real-time PCR and western blot were used to evaluate the expression levels of ACC1 in 10 paired ovarian cancer and adjacent non-cancer tissues. MTS and EDU assays were used to determine the proliferation of ovarian cancer cells when ACC1 was knocked-down by siRNA. Flow cytometry was used to determine the apoptosis of ovarian cancer cells when ACC1 was knocked-down by siRNA. Results The expression of ACC1 is elevated ovarian cancer tissues at both mRNA and protein levels( P <0.01). The proliferation of ovarian cancer cells were significantly decreased when ACC1 was knocked-down( P <0.01). No significant change of the apoptosis of ovarian cancer cells was observed when ACC1 was knocked-down in ovarian cancer cells( P >0.05). Conclusion ACC1 is over-expressed in human ovarian cancer tissues, which promotes the proliferation of ovarian cancer cells, indicating ACC1 as a potential therapeutic target for the treatment of ovarian cancer.
