摘  要:采用光谱法、热力学理论以及盐效应作用等手段,研究了灿烂甲酚蓝(BCB)与DNA的结合模式,结果表明,BCB可与DNA形成复合物发生荧光淬灭,二者淬灭常数随温度升高而减小,说明DNA对BCB淬灭机制为静态淬灭;ΔG<0,表明二者的相互作用可自发进行;且ΔS>0,ΔH<0,说明BCB和DNA之间主要作用力为氢键和疏水作用力,NaCl和NaH2PO4的加入对BCB-DNA体系的荧光强度没有影响,故二者之间不存在静电作用;BCB浓度增大,DNA-盐酸小檗碱(BR)体系的荧光强度减弱,说明BCB与DNA的结合模式类似于BR与DNA的结合,为嵌插结合.Spectroscopic methods,thermodynamics theory and salt effect were adopted to study the binding mode between brilliant cresyl blue(BCB)and DNA.The results showed that the fluorescence of BCB was quenched remarkably due to the formation of BCB-DNA complex.Furthermore,its quenching mechanism was static quenching,because the quenching constant(Ka)of BCB with DNA was decreased with the increase of temperature.ΔG was negative,andΔS>0,ΔH<0,indicating that interaction between BCB and DNA were spontaneously bounded by hydrogen bonding and hydrophobic interaction force.The addition of NaCl and NaH 2PO 4 had no effect on the fluorescence intensity of BCB-DNA system.There was no electrostatic interaction between the BCB and DNA.With the increase of the BCB concentration,the fluorescence intensity of DNA-Berberine(BR)system was weakened,indicating that the binding mode of BCB to DNA was similar to the binding of BR to DNA,which was insert mode.
