Effects of Tangtong Formula on Expressions of PARP and NF-κB in Sciatic Nerve of Diabetic Peripheral Neuropathy Rats

摘  要:目的:观察中药糖痛方对DPN大鼠坐骨神经PARP及NF-κB表达的影响,并探讨其保护作用机制。方法:SD雄性大鼠75只,随机选取15只为正常组,余60只大鼠予一次性腹腔注射STZ溶液(60 mg/kg)制备糖尿病大鼠模型。4周后结合缺血再灌注制备糖尿病周围神经病变(DPN)大鼠模型,DPN造模成功后随机分为模型组、糖痛方小、中、大剂量组,每组15只,糖痛方小、中、大剂量组按成人6.25倍、12.5倍、25倍剂量分别给予糖痛方9.17、18.33、36.67 g/kg灌胃,1次/d,连续给药8周,正常组和模型组灌胃等体积蒸馏水。实验结束后,应用Western Blot法及实时荧光定量PCR检测各组大鼠坐骨神经PARP及NF-κB的表达。结果:与正常组大鼠比较,模型组大鼠坐骨神经PARP-1、PARP-2蛋白和mRNA相对表达均升高;模型组NF-κB mRNA相对表达明显高于正常组(P<0.01),差异具有统计学意义。经糖痛方灌胃给药8周后,与模型组比较,各中药组PARP-1、PARP-2蛋白相对表达有不同程度下降(P>0.05),且下降程度与糖痛方剂量呈正相关,PARP-1、PARP-2 mRNA相对表达除糖痛方小剂量组外,糖痛方中、大剂量组PARP-1、PARP-2mRNA表达以剂量依赖性方式呈现出不同程度下降趋势(P>0.05),与此同时,各中药组NF-κB mRNA相对表达也有不同程度下降。结论:中药糖痛方可通过下调DPN大鼠坐骨神经PARP及NF-κB的表达水平,抑制周围神经损伤的炎症反应,从而起到防治DPN作用。Objective:To observe the effect of Tangtong Formula on the expressions of PARP and NF-κB in sciatic nerve of diabetic peripheral neuropathy rats and meanwhile explore its protective mechanism. Methods:Among Seventy-five healthy SD male rats,15 were selected randomly as the normal group and the other 60 rats were treated with intraperitoneal injection of streptozotocin(STZ)60 mg/kg to build type 2 diabetic mice model. After feeding for 4 weeks,combining ischemia-reperfusion model to make diabetic peripheral neuropathy rats model. After the models establishment,the 60 DPN model rats were randomly divided into 4 groups:model group,tangtong formula low-dose group,Tangtong Formula medium-dose group and Tangtong Formula high-dose group,15 rats each group. The Tangtong Formula low,medium and high-dose groups were given Tangtong Formula 9.17,18.33,36.67 g/kg with the dose of 6.25,12.5,25 times of the adults. The normal group and the model group were given the equal water,once per day for 8 weeks. At the end of the experiment, the expressions of PARP and NF-κB in the sciatic nerve of each group were detected by Western blot and quantitative real-time RT-PCR methods. Results:Compared with the normal group,the protein and mRNA expression of PARP-1 and PARP-2 in the model group were increased and the relative expression of NF-κB mRNA in the model group was obviously increased(P<0.01). The difference was significant. After treating with Tangtong Formula for 8 weeks,the relative expressions of PARP-1 and PARP-2 protein in each Tangtong Formula treatment group were decreased in varying degrees when compared with those of the model group(P>0.05)and the effect was positively correlated with the dose of Tangtong Formula. Except for the Tangtong Formula low dose group,the relative expressions of PARP-1 and PARP-2 mRNA were decreased in the Tangtong Formula medium and high-dose groups(P>0.05),which showed a dose dependent manner.
