摘 要:目的:探讨化瘀祛痰方对高脂血症大鼠sd-LDL的影响和机制。方法:80只大鼠随机分为正常组、模型组、化瘀祛痰组和辛伐他汀组,每组20只,正常组给予普通饲料喂养12周,其余3组给予高脂饲料喂养,诱导高脂血症。共喂养12周。造模结束后,化瘀祛痰组和辛伐他汀组分别给予化瘀祛痰方和辛伐他汀灌胃。全自动生化分析仪检测血清TG、TC、HDL-C、LDL-C含量,Lipoprint脂蛋白分析系统检测LDL颗粒大小,并计算sd-LDL占总LDL百分比,实时荧光定量PCR技术和Western blotting技术分别检测肝组织中LPL、LCAT的mRNA和蛋白表达。结果:与正常组比较,模型组大鼠血清TG、TC、LDL-C水平升高,HDL-C水平降低,LDL平均颗粒直径较小,sd-LDL浓度比升高,大鼠肝脏LPL、LCATmRNA及蛋白表达均降低(P<0.01,P<0.05)。与模型组比较,化瘀祛痰组和辛伐他汀组大鼠血清TG、TC水平降低,LDL平均颗粒直径变大,sd-LDL浓度比降低,LPL、LCAT mRNA及蛋白表达均升高(P<0.01,P<0.05)。结论:化瘀祛痰方可以降低高脂血症大鼠sd-LDL浓度比,从而降低了AS发生的风险。Objective:To explore the effect and mechanism of Huayu Qutan Prescription on sd-LDL in rats with hyperlipidemia.Methods:Eighty rats were randomly divided into normal group, model group, Huayu Qutan group and simvastatin group, with 20 rats in each group.The normal diet was given normal diet while the other groups were fed high fat food.Huayu Qutan group and simvastatin group were given Huayu Qutan Prescription and simvastatin for gastric perfusion, respectively.The contents of serum TG, TC, HDL-C and LDL-C were detected by automatic biochemical analyzer.The Lipoprint lipoprotein analysis system detected the size of LDL particles and calculated the percentage of sd-LDL in total LDL. The mRNA and protein expressions of LPL and LCAT in liver tissues were detected by real-time fluorescence quantitative PCR and Western blotting.Results:Compared with normal group,the levelsof TG,TC and LDL-C in model group were increased while HDL-C decreased, and LDL mean particle diameter was smaller while sd-LDL percentage increased. LPL and LCAT mRNA and protein expressions in liver decreased(P<0.01,P<0.05).Compared with the model group, the levels of TG and TC in the Huayu Qutan group and simvastatin group decreased. The average particle diameter of LDL increased, the percentage of sd-LDL decreasedand the expressions of LPL and LCAT mRNA and protein increased(P<0.01,P<0.05).Conclusion:Huayu Qutan Prescription can reduce the concentration percentage of sd-LDL in rats with hyperlipidemia,which can reduce the risk of AS.
