摘 要:目的探讨膀胱尿路上皮癌组织中Gankyrin、CyclinD1及HER-2的表达及其临床病理特征的关系。方法应用组织芯片技术和免疫组化法,检测Gankyrin、CyclinD1及HER-2蛋白在87例浸润性膀胱尿路上皮癌和42例慢性膀胱炎黏膜组织中的表达情况,并分析三者的表达与临床病理特征的关系。结果42例慢性膀胱炎黏膜组织Gankyrin、CyclinD1呈低表达,87例浸润性膀胱尿路上皮癌组织中Gankyrin、CyclinD1的阳性率为71.3%和56.3%,差异有统计学意义(P<0.01)。Gankyrin、CyclinD1在膀胱尿路上皮癌中的阳性率随组织学分级的升高而升高(P=0.001、P=0.004),且两者的表达呈正相关。Gankyrin、CyclinD1的表达与患者的年龄、性别以及有无肌层浸润无关(P>0.05)。HER-2在膀胱尿路上皮癌组织中的阳性率为13.8%,在慢性膀胱炎黏膜组织中无表达,与患者性别、年龄、组织学分级及有无肌层浸润无关(P>0.05)。结论Gankyrin、CyclinD1在膀胱癌中高表达且与其组织学分级显著相关,且两者的表达呈正相关,有望成为膀胱尿路上皮癌的潜在生物学标志物。Objective To investigate the expression of Gankyrin, Cyclin D1 and HER-2 in bladder urothelial carcinoma tissue and its relationship with clinicopathological features. Methods Tissue microarraycontaining specimens from 87 patients with bladder urothelial carcinoma and 42 normal controls was constructed and immunohistochemistry was applied to examine the expression of Gankyrin, Cyclin D1 and HER-2, and the correlations between the expression of Gankyrin, Cyclin D1 and HER-2 and clinicopathological characteristics were analyzed. Results Immunohistochemical detection showed normal bladder urothelium from42 control cases were weak protein expression of Gankyrin and Cyclin D1, The positive expression rates of Gankyrin and Cyclin D1 in 87 cases of bladder urothelial carcinoma were 71.3% and 56.3%, the difference was statistically significant(P<0.01). The expression of Gankyrin and Cyclin D1 in bladder urothelial carcinoma was positively associated with higher histological grade(P=0.001, P=0.004), and the expression of both was positively correlated. The Gankyrin and Cyclin D1 expression level had no relationship with age, gender and presence or absence of myometrial invasion(P >0.05). The expression rate of HER-2 in bladder urothelial carcinoma was 13.8%, it was not expressed in normal control tissues, it was not related to sex, age, histological grade and presence or absence of myometrial invasion(P >0.05). Conclusion Gankyrin and Cyclin D1 are high expressed in bladder urothelial carcinoma and are significantly correlated with histological grade, and their expression is positively correlated, indicating that it might be a potential bio-marker for bladder urothelial carcinoma.
