叶下珠多酚对兔离体肠运动的舒张作用

摘　　要：目的:本试验以兔离体肠平滑肌为研究对象,考察叶下珠多酚对离体肠肌收缩的影响,并探讨其作用机制。方法:利用BL-420E生物机能系统,观察叶下珠多酚对离体肠肌自发性收缩的影响;观察叶下珠多酚对工具药氯化钡、酚妥拉明及组胺作用下离体肠肌活动的影响,并记录收缩频率和振幅。结果:叶下珠多酚可抑制兔离体肠肌自发性收缩的振幅,且呈剂量依赖性;叶下珠多酚可显著或极显著抑制由氯化钡、酚妥拉明及组胺引起的离体肠肌的挛缩(P<0.05或P<0.01)。结论:推测叶下珠多酚的抗腹泻作用与抑制小肠蠕动有关,叶下珠多酚抑制肠肌运动可能与加快肠上皮细胞对钙离子的释放、促进胞外钙离子内流的H1受体受阻及激动α受体有关。Objective:To clear the effects of polyphenols from Phyllanthus urinaria on contraction of intestinal smooth muscle of rabbit in vitro. Method:The experiments were performed in isolated intestinal smooth muscle of rabbits. By analyzing the frequency and amplitude of contraction,the inhibition of polyphenols from Phyllanthus urinaria on normal intestinal smooth muscle contraction and histamine,phentolamine and barium chloride(BaCl2)-treated intestinal muscle contraction were compared. Result:The results showed that polyphenols from Phyllanthus urinaria significantly inhibited the frequency and amplitude of spontaneous contraction of isolated intestinal smooth muscle with dose-dependent. Polyphenols from Phyllanthus urinaria inhibited intestinal smooth muscle contraction caused by phentolamine,BaCl2 and histamine. Conclusion:It was speculated that the anti-diarrhea effect was related to the inhibition of intestinal peristalsis. Polyphenols from Phyllanthus urinaria inhibiting intestinal muscle movement may be related to promoting the release of calcium ions from intestinal epithelial cells,blocking H1 receptors and stimulating α receptors.
