摘 要:目的探讨结直肠癌中赖氨酰氧化酶样蛋白-2(LOXL-2)的表达及其与临床参数及预后的关系。方法采用荧光定量PCR和免疫组化染色技术检测结直肠癌组织和正常肠黏膜组织中LOXL-2 mRNA和蛋白的表达情况,分析LOXL-2的表达与年龄、淋巴结转移数目、TNM分期、肿瘤分级及预后的相关性。结果 LOXL-2在结直肠癌组织和淋巴结转移癌组织中的表达水平显著高于正常肠黏膜组织(P<0.01)。LOXL-2表达水平与结直肠癌患者的临床分期和淋巴结转移相关(P<0.05)。LOXL-2高表达的患者其3年和5年的总体生存率(OS)均显著低于LOXL-2低表达患者,差异有统计学意义(P=0.037)。LOXL-2和淋巴结转移是OS(HR=3.368,P=0.007;HR=2.457,P=0.013)的独立预后因素。结论 LOXL-2的表达与结直肠癌患者的淋巴结转移及肿瘤分期密切相关,结直肠癌组织中LOXL-2高表达为结直肠癌患者预后差的风险因素。Objective To investigate the expression of lysinylase-like protein-2 ( LOXL-2) in colorectal cancer and its correlation with clinical indicators and prognosis. Methods The expression of LOXL-2 mRNA and protein in colorectal cancer tissues and normal intestinal mucosal tissues was assessed by fluorescence quantitative PCR and immunohistochemical staining. The correlations between LOXL-2 expression and age,number of lymph node metastases, TNM stage,tumor grade and prognosis were analyzed. Results The expression levels of LOXL-2 in colorectal cancer tissues and lymph node metastatic carcinoma tissues were significantly higher than that in normal intestinal mucosal tissues ( P < 0. 01). The expression level of LOXL-2 was associated with clinical staging and lymph node metastasis in patients with colorectal cancer ( P < 0. 05). The OS of patients with high expression of LOXL-2 for 3 and 5 years were significantly lower than those with low expression of LOXL-2 ( P = 0. 037). LOXL-2 and lymph node metastasis were independent prognostic factors for OS ( HR = 3. 368,P = 0. 007;HR = 2. 457,P = 0. 013). Conclusion The expression of LOXL-2 is closely related to lymph node metastasis and tumor staging in patients with colorectal cancer. High expression of LOXL-2 in colorectal cancer tissues is a risk factor for poor prognosis in colorectal cancer patients.
