摘 要:目的:探究BTF3基因对人膀胱癌细胞株T24生物学功能的影响。方法:通过慢病毒感染的方法,构建BTF3基因低表达的T24细胞株。通过体外平板克隆实验检测克隆形成,MTT检测细胞增殖,流式细胞术检测细胞凋亡。结果:RT-PCR和Western blotting证实BTF3低表达的T24细胞株构建成功。与对照组的细胞相比,敲低BTF3的表达能够抑制T24细胞克隆形成和细胞增殖,促进细胞凋亡。结论:我们的研究结果证明了BTF3在人膀胱癌细胞株T24中的重要性,提示BTF3不仅可能与膀胱癌的发生有关,而且可能成为膀胱癌潜在的治疗靶点和生物标记物。Objective:To explore the effect of BTF3 gene on the biological function of human bladder cancer cell line T24.Methods:A T24 cell line with low expression of the BTF3 gene was constructed by a method of lentivirus infection.Colony formation was detected by in vitro plate cloning assay.Cell proliferation was detected by MTT,and apoptosis was detected by flow cytometry.Results:RT-PCR and western blotting confirmed that the T24 cell line with low expression of BTF3 was successfully constructed.Compared with the cells of the control group,knockdown of BTF3 expression can inhibit the colony formation,cell proliferation and promote apoptosis of T24 cells.Conclusion:Our results demonstrate the importance of BTF3 in human bladder cancer cell line T24,suggesting that BTF3 may not only be involved in the development of bladder cancer,but may also be a potential therapeutic target and biomarker for bladder cancer.
