摘　　要：目的:研究蛋白激酶CK2β、人附睾分泌蛋白-4(HE-4)、细胞信号调节蛋白激酶1/2(ERK1/2)在卵巢性索间质瘤模型大鼠中的表达水平。方法:选取40只健康Wistar大鼠,随机分为对照组和模型组,各20只,建立卵巢性索间质瘤模型,采用Western blot检测蛋白激酶CK2β、HE-4、ERK1/2蛋白表达水平,荧光定量RT-PCR技术检测CK2β、HE-4、ERK1/2基因水平。结果:模型组大鼠蛋白激酶CK2β水平显著高于对照组大鼠,模型组大鼠CK2βmRNA水平显著高于对照组大鼠,具有统计学差异(P<0.05)。模型组大鼠HE-4水平显著高于对照组大鼠,模型组大鼠HE-4mRNA水平显著高于对照组大鼠,具有统计学差异(P<0.05)。模型组大鼠ERK1/2水平显著高于对照组大鼠,模型组大鼠ERK1/2 mRNA水平显著高于对照组大鼠,具有统计学差异(P<0.05)。蛋白激酶CK2β、HE-4、ERK1/2的敏感度、特异度、准确性比较存在差异,ERK1/2单独检测的敏感度、特异度、准确性显著高于其他两项,具有统计学差异(P<0.05);三项因子联合检测的敏感度、特异度、准确性显著高于三项单独检测,具有统计学差异(P<0.05)。结论:蛋白激酶CK2β、HE-4、ERK1/2在卵巢性索间质瘤模型大鼠中显示高表达,其三项联合检测的诊断效能较高,对卵巢性索间质瘤患者的治疗和预后具有重要的参考价值。Objective:To study expression levels of protein kinase CK2 β,HE-4 and ERK1/2 in rats with specific sex cord-stromal ovarian tumor.Methods:40 healthy Wistar mice were randomly divided into the control group and the model group,each 20.The specific sex cord interstitial ovarian cancer model was established.The expression level of protein kinase CK2β,HE-4,ERK1/2 protein was detected by Western blot,and the gene level of CK2β,HE-4 and ERK1/2 was detected by the fluorescence quantitative RT-PCR technique.Results:The level of protein kinase CK2β in the model group was significantly higher than that in the control group,and the level of CK2β mRNA in the model group was significantly higher than that in the control group,with statistical difference(P<0.05).The level of HE-4 in the model group was significantly higher than that in the control group,and the level of HE-4 mRNA in the model group was significantly higher than that in the control group,with a statistically significant difference(P<0.05).The level of ERK1/2 in the model group was significantly higher than that in the control group,and the level of ERK1/2 mRNA in the model group was significantly higher than that in the control group,with a statistically significant difference(P<0.05).The sensitivity,specificity and accuracy of protein kinase CK2β,HE-4 and ERK1/2 were different.The sensitivity,specificity and accuracy of ERK1/2 were significantly higher than those of the other two items,with statistical difference( P < 0. 05),and the sensitivity,specificity and accuracy of the three factors combined detection were significantly higher than that of three individual tests. It has statistical significance( P < 0. 05). Conclusion: The protein kinase CK2β,HE-4 and ERK1/2 are highly expressed in the rats model of specific sex stromal ovarian cancer,and the diagnostic efficiency of the three joint tests is very high. It has important reference value for the treatment and prognosis of the patients with specific sex cord-stromal ovarian tumor.
