摘 要:目的研究血小板反应蛋白-4(TSP-4)在大鼠口面部肌源性疼痛中的作用。方法SD大鼠分为结扎组(L组,结扎咬肌浅层肌腱)和假手术组(S组,仅暴露肌腱),每组6只。术前及术后3、7、10、14d测疼痛阈值。术后14d收集三叉神经脊束核尾侧亚核及颈段第二脊髓背侧角(V_C/C2),测TSP-4mRNA表达水平。SD大鼠肌腱结扎后7d,咬肌局部注射TSP-4抗体(A组,抗体组)或PBS(C组,对照组),每组6只。注射前及注射后2、4、6、12h测疼痛阈值。结果与S组比较,L组疼痛阈值在术后3、7、10、14d明显降低(P<0.05),TSP-4mRNA表达水平在术后14d显著上调(P<0.05)。与C组比较,A组疼痛阈值在注射后4、6h明显升高(P<0.05)。结论TSP-4参与大鼠口面部肌源性疼痛的发生发展。Objective To study the role of thrombospondin-4(TSP-4) in rats with myogenic orofacial pain. Methods Twelve SD rats were divided into ligation group(the anterior superficial part of masseter muscle was ligated) and sham group(without ligation). The mechanical threshold was assessed before surgery and 3, 7, 10, 14 days after surgery. TSP-4 mRNA levels in trigeminal spinal subnucleus caudalis and C1-C2 spinal cervical dorsal horn(V_C/C2) were measured 14 days after surgery. Twelve rats of ligation group were divided into antibody group(TSP-4 antibody injection) and control group(PBS injection). The mechanical threshold was assessed before injection and 2, 4,6, 12 hours after injection. Results Compared with sham group, the mechanical threshold was significantly decreased and maintained throughout 14 days observation in ligation group(P<0.05). The levels of TSP-4 m RNA were upregulated significantly 14 days after surgery(P<0.05). Compared with control group, the mechanical threshold was increased at 4 and 6 hours after TSP-4 antibody injection(P<0.05). Conclusion TSP-4 is involved in the pathogenesis of myogenic orofacial pain of rats.
