摘 要:目的:研究血当归总黄酮提取工艺及其体外抗氧化活性。方法:采用超声法提取血当归总黄酮,在510nm处测定总黄酮的含量,并通过单因素实验和L9(33)正交设计对提取工艺进行优化。并以抗坏血酸为阳性对照,探讨了其对1,1-二苯基-2-三硝基苯肼自由基(DPPH·),2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS^+·)及羟自由基(·OH)的清除能力。结果:血当归总黄酮提取的最佳条件为料液比1∶30g·mL^-1,乙醇浓度65%,提取时间45min,提取血当归总黄酮的含量可达90.05mg·g^-1。此条件下得到的血当归总黄酮提取物清除DPPH·,ABTS^+·和·OH的能力良好,尤其对ABTS+·清除能力最优,其IC50值分别为11.62、1.83、136.12mg·L^-1。结论:优化的提取工艺稳定、合理,血当归总黄酮有较好的体外抗氧化活性并呈现出良好的量效关系。Objective To study extraction technology of total flavonoids from the Polygonum runcinatum and its antioxidant activities in vitro. Methods Thetotal1flavonoids were extracted byultrasonicfrom the P. runcinatum, the content of total flavonoids was determined at 510 nm, and the extraction technology was optimized with single factor experiment and L 9 (3 3) orthogonal design.Withthe ascorbic acid as the contrast, its scavenging activities of DPPH·, ABTS +·,·OHwerediscussed. Results the optimum condition of totalflavonoids extraction: Solid-liquid ratio was1∶ 30 g·mL^-1 , ethanol concentration was 65%, and extraction time was 45 min, the content of total flavonoids extracted was up to 90.05 mg·g ^-1 .The totalflavonoids extracted from the P.runcinatumunder these conditions had good ability to scavenge several free radicals, especially ABTS +·, the IC 50 of DPPH·, ABTS^+·and·OH were 11.62, 1.83, 136.12 mg·L ^-1 . Conclusion The optimized extraction technology was stable and reasonable. The total flavonoids of P.runcinatum had better antioxidant activity and show a good dose-effect relationship.
