摘  要:目的探讨透明质酸(HA)酶对人牙周膜成纤维细胞(HPDLFs)增殖的影响及作用机制。方法从健康人前磨牙中刮取根中部1/3的牙周膜组织,分离HPDLFs。干预方式:(1)采用15 U/ml、75 U/ml和150 U/ml的HA酶干预HPDLFs,对照组加入同剂量的PBS溶液。(2)采用2μg/ml的CD44抗体干预HPDLFs,阴性对照为2μg/ml的Ig G干预,对照组加入同剂量的PBS溶液。于干预的第1、2、3、5、7、9、11天采用MTT法检测细胞增殖。采用实时定量PCR检测75 U/ml的HA酶或CD44抗体干预HPDLFs 24 h后透明质酸合成酶(HASs)、HA酶(HYALs)和CD44 mRNA的表达。结果与对照组相比,15 U/ml、75 U/ml和150 U/ml的HA酶均呈剂量依赖式抑制HPDLFs的增殖(P <0. 05)。与对照组相比,CD44抗体可显著抑制HPDLFs的增殖(P <0. 05)。与对照组相比,75 U/ml的HA酶或CD44抗体均可显著抑制HAS1、HAS2和HAS3 mRNA的表达,增加HYAL 1、HYAL 2和HYAL 3 mRNA的表达(P <0. 05),但对CD44 mRNA的表达无显著影响。结论 CD44可介导HA酶抑制HPDLFs的增殖。Objective To investigate the effect and its mechanism of hyaluronidase on the proliferation of human periodontal ligament fibroblasts (HPDLFs). Methods 1/3 of the periodontal tissue in the middle of the root from the healthy premolars was scraped and HPDLFs were isolated from it. Means of intervention:① HPDLFs were intervened with hyaluronidase of 15 U/ml, 75 U/ml and 150 U/ml, and the same dose of PBS solution was added to the control group.②HPDLFs were intervened with 2 μg/ml of CD44 antibody, the negative control was intervened with 2 μg/ml of IgG, and the control group was added with the same dose of PBS solution. Cell proliferation was measured by MTT assay on days 1, 2, 3, 5, 7, 9, and 11 of the intervention. Expressions of hyaluronan synthase (HASs), hyaluronidase (HYALs) and CD44 mRNA after 24 hours of HPDLFs intervention by 75 U/ml hyaluronidase or CD44 antibody were detected by Real-time quantitative PCR. Results Compared with the control group, the hyaluronidases of 15 U/ml, 75 U/ml and 150 U/ml inhibited the proliferation of HPDLFs in a dose-dependent manner ( P <0.05). Compared with the control group, CD44 antibody significantly inhibited the proliferation of HPDLFs ( P <0.05). Compared with the control group, 75 U/ml of HA enzyme or CD44 antibody significantly inhibited the expressions of HAS1, HAS2 and HAS3 mRNA, and increased the expressions of HYAL 1, HYAL 2 and HYAL 3 mRNA ( P <0.05), but there was no significant effect on CD44 mRNA expression. Conclusion Hyaluronidase inhibits proliferation of HPDLFs by mediating CD44.
