磷酸化Tyr951信号蛋白在糖尿病大鼠创面组织内皮细胞迁移中的作用

摘  要:目的观察糖尿病大鼠创面组织内皮细胞迁移信号通路中磷酸化Tyr951表达的变化规律。方法SD大鼠分为糖尿病组和非糖尿病对照组,建立深Ⅱ度烫伤模型,透明胶纸描计计算大鼠创面愈合率,于烫伤后0,1,3,7,10,14,21d,ELISA法检测创面组织中F-Actin的水平,免疫组织化学方法检测创面组织中磷酸化Tyr951的表达。结果与非糖尿病组相比,糖尿病组创面组织中磷酸化Tyr951表达水平显著降低(P<0.05),同时F-Actin表达水平也显著降低(P<0.05)。结论糖尿病大鼠创面组织内皮细胞迁移障碍与磷酸化Tyr951信号蛋白表达减少有关。OBJECTIVE To observe the expression of phosphorylated Tyr951 in endothelial cell migration signaling pathway in diabetic rats. METHODS SD rats were divided into diabetic group and non-diabetic control group,and then the rats of the two groups were produced the deep partial thickness scald on the dorsal skin.The rate of wound healing in the two groups was calculated by using a transparent rubber paper plotter.The expressions of F-Actin in wound tissue were detected by enzyme-linked immunosorbent assay and the contents of phosphorylated Tyr951 in wound tissue were determined with immunohistochemical method on post scald day 0,1,3,7,10,14,21. RESULTS Compared with the non-diabetic group,the expression of phosphorylated Tyr951 in the wound tissue of the diabetic group was significantly lower than that in the non-diabetic group ( P <0.05),and the expression of F-Actin was also significantly decreased ( P <0.05). CONCLUSION The dysfunction of endothelial cell migration in diabetic rats was related to the decrease of phospho-Tyr951 signal protein expression.
