摘　　要：目的探讨Mortalin蛋白过表达在胃腺癌(GAC)临床预后评估中的意义。方法应用免疫荧光染色法观察Mortalin蛋白在人胃癌BGC-823细胞中的表达定位;对TCGA数据库和Oncomine数据库中的样本数据进行分析以确定Mortalin在GAC中的表达水平;采用免疫组化检测Mortalin蛋白的表达,并分析其与临床病理参数之间的关系;分析Mortalin蛋白表达水平与GAC患者预后的关系。结果 Mortalin蛋白主要表达于GAC细胞质中,且在GAC中过表达。GAC组织中Mortalin阳性率、强阳性率均明显高于胃上皮内瘤变和正常胃黏膜组织。Mortalin蛋白表达水平与GAC分化程度、浸润深度、淋巴结转移密切相关。Mortalin蛋白高表达的GAC患者生存期更短。结论 Mortalin蛋白表达水平与GAC的发生、发展及预后密切相关。Mortalin蛋白过表达可成为GAC患者不良预后的重要指标之一。Objective To investigate the significance of Mortalin protein over-expression on prognostic evaluation of gastric adenocarcinoma(GAC). Methods The localization of Mortalin protein in BGC-823 cells was observed by immunofluorescence(IF) staining. The sample data in TCGA database and Oncomine database were analyzed to determine the expression of Mortalin in GAC. The expression of Mortalin was detected by immunohistochemistry and the relationship between Mortalin and clinicopathologic parameters was analyzed. The relationship between Mortalin protein expression level and the prognosis of GAC patients was analyzed by TCGA database and immunohistochemical results.Results(1)Mortalin protein was mainly located in GAC cytoplasm and overexpressed in GAC.(2) The positive rate and strongly positive rate of Mortalin were significantly higher than those either in intraepithelial neoplasia or in normalcarcinoma tissues. Mortalin expression was significantly correlated with the differentiation, infiltration depth, LN metastasis of GAC.(3)The prognosis of GAC patients with high Mortalin protein expression was poor. Conclusions Mortalin expression level is closely related to the progression and prognosis of GAC. High level of Mortalin expression might partially be used as an important poor prognostic indicator for GAC.
