摘 要:本试验旨在利用同位素标记法研究瘤胃微生物对蛋氨酸(Met)的利用及Met对微生物氮流转的影响。试验以玉米-豆粕型精料(0.5 g)和玉米-秸秆型饲粮(0.5 g)为发酵底物,采用单因子试验设计,设试验组和对照组。试验组添加0. 33μmol同位素标记的碳13蛋氨酸(13C-M et,以0.18 mL溶液的形式添加到体外培养底物中),对照组添加等量蒸馏水,探究M et对体外发酵pH和氨态氮(NH3-N)、挥发性脂肪酸(VFA)、微生物氮、微生物M et含量以及微生物13C丰度的影响。结果表明:1)添加M et能极显著提高发酵液的pH(P<0.01);2)添加M et能极显著提高微生物13C的丰度(P<0.01);3)添加M et对NH3-N、VFA、微生物氮、微生物M et含量均没有显著影响(P>0.05)。由此可见,瘤胃微生物可有效利用Met,Met对发酵特性有一定的影响,但不影响微生物氮的流转。The aim of this study was to investigate the utilization of methionine(Met) by rumen microorganisms and effects of Met on microbial nitrogen transfer. Corn-soybean meal concentrate and(0.5 g) and cornstraw diet(0.5 g) were used for fermentation substrates,the study was conducted as one-factor block experimental design,and set up test group and control group. The test group was supplemented with 0.33 μmol isotope labeling 13 C methionine(13 C-Met,addition of 0.18 mL solution to in vitro culture substrate),and the control group was supplemented with equivalent distilled water,to explore the effects of Met on in vitro fermentation pH,the contents of ammonia nitrogen(NH3-N),volatile fatty acid(VFA),microbial nitrogen and microbial Met and the abundance of 13 C in microbial. The results showed as follows: 1) adding Met could significantly increase the pH of fermentation liquid(P<0.01);2) adding Met could significantly increase the abundance of 13 C in microorganisms(P < 0. 01);3) adding Met had no significant effects on the contents of NH3-N,VFA,microbial nitrogen and microbial Met(P>0.05). Therefore,it can be inferred that rumen microorganisms can effectively utilize Met,Met has some influence on fermentation characteristics,but do not affect the flow of microbial nitrogen.
