摘 要:本试验旨在采用代谢组学技术研究芦笋皮或象草对肉牛血清代谢物的影响。选择健康、年龄相近,体重为(236.50±34.00) kg的安格斯肉牛10头,随机分为2组,每组5个重复,每个重复1头肉牛。2组分别饲喂以芦笋皮或象草作为粗饲料配制的全混合日粮(TMR)。预试期15 d,正试期45 d。正试期最后1天时,于晨饲前采尾静脉血,分离血清,利用气相色谱-质谱(GC-MS)联用技术检测血清代谢物,主成分分析(PCA)展示代谢物的分布情况,筛选并鉴定差异代谢物,分析涉及的代谢通路。结果表明:2组肉牛血清中共检出16个差异代谢物。与象草组相比,芦笋皮组血清中缩二脲、β-谷氨酸、环亮氨酸、脱氧葡萄糖等12个代谢物上调,L-半胱氨酸、对异丙苯甲酸、N-乙酰色氨酸和异亮氨酸下调,涉及到丙氨酸、天冬氨酸和谷氨酸代谢,精氨酸和脯氨酸代谢,缬氨酸、亮氨酸和异亮氨酸生物合成以及柠檬酸循环等7个通路。由此可见,在本试验条件下,肉牛饲喂芦笋皮比象草更有利于营养物质代谢。This study was conducted to investigate the effects of asparagus peel or napiergrass on serum metabolites of cattle based on metabonomics technique. Ten healthy Angus beef cattle [(236.50±34.00) kg of body weight]with similar ages were randomly divided into 2 groups with 5 replicates per group and 1 cattle per replicate.Cattle in the two group were fed the total mixed rations(TMR) with asparagus peel or napiergrass as roughage. The pre-experimental period lasted for 15 days,and the experimental period lasted for 45 days. On the last day of experiment,the blood samples were collected from tail venous before morning feeding,and then serum was isolated,and serum metabolites were detected by gas chromatography-mass spectrometry(GC-MS)technique,using principal component analysis(PCA) showed the distribution of metabolites,screened and identified the metabolites,and analysis the metabolic pathways. The results showed that 16 different metabolites were obtained in two groups. Twelve metabolites of asparagus peel group were up-regulated compared with napiergrass group including biuret,β-glutamic acid,cycloleucine and 2-deoxy-D-glucose,etc,but L-cysteine,cumic acid,N-acetyltryptophan and isoleucine were down-regulated,these metabolites were mainly related to 7 pathways including alanine,aspartate and glutamate metabolism,arginine and proline metabolism,valine,leucine and isoleucine biosynthesis and citrate cycle,etc. In conclusion,under the conditions of this experiment,feeding cattle with asparagus peel is more beneficial to nutrient metabolism than napiergrass.
