摘  要:目的从采自广西北部湾的泥样和植物中分离海洋真菌,筛选具有抗菌活性的真菌菌株,分离鉴定活性化合物。方法采用普通稀释法从样品中分离菌株,对次级代谢产物进行抑菌活性测试,以活性为导向分离抗菌活性化合物,并通过波谱方法确定化合物的结构。结果从采自广西北部湾的2个海泥和8个植物样品中分离72株真菌,筛选得到对香蕉枯萎病菌(Fusarium oxysporum f. sp cubense)、白色念珠菌(Candida albicans)、金黄色葡萄球菌(Staphylococcus aureus)和耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)等致病菌有抑菌作用的活性菌株13株,并从其中的1株真菌MDCW-15次级代谢产物中分离鉴定了烟曲霉素。抑菌活性测试结果显示,该化合物对白色念珠菌的最小抑菌浓度为64 μg/ml。结论从广西北部湾分离的药用活性真菌资源具有深入研究的价值。Objective To obtain bioactive fungal strains with antimicrobial activity from samples and plants collected in the Guangxi Beibu Gulf region,and identify the active metabolites. Methods The isolation of fungal strains was performed by the dilution method. The antimicrobial activity was assayed for the fungal strains using culture extracts to obtain bioactive fungal strains. The separation of antibacterial metabolites was carried out in a bioassay-guided manner. The structure of obtained compound was identified by the spectroscopic method. Results Seventy- two fungal strains were isolated from the soil and plant samples collected in the Guangxi Beibu Gulf region. Among them,13 strains exhibited antimicrobial activities on Fusarium oxysporum f. sp cubense,Candida albicans,Staphylococcus aureus or methicillin- resistant Staphylococcus aureus(MRSA). Fumagillin was isolated and identified from the secondary metabolites of one of the 13 fungal strains,MDCW-15. In the present bioassay,fumagillin displayed antifungal activity against C. albicans with the minimum inhibitory concentration(MIC)of 64.0 μg/ml. Conclusion Fungi from the Guangxi Beibu Gulf region are potential resources of antimicrobial fungal strains,whose active metabolites is worth further investigation.
