食管癌Survivin乙酰化水平与PCAF表达的相关性及其临床意义

摘　　要：【目的】探讨食管癌组织中Survivin蛋白的乙酰化水平和P300/CBP相关因子(PCAF)的表达与临床病理特征之间的关系。【方法】采用免疫共沉淀和Western blot检测90例食管癌和癌旁正常食管上皮组织中Survivin蛋白乙酰化水平;免疫组织化学染色法检测PCAF在食管癌组织中的表达水平;分析Survivin蛋白乙酰化水平与患者临床病理特征的关系及PCAF蛋白表达的相关性。【结果】食管癌组织中Survivin蛋白乙酰化率为(72.67±1.25)%,显著高于癌旁正常食管组织中的(11.18±2.16)%(P<0.05);食管癌组织中Survivin蛋白乙酰化水平与患者的TNM分期、组织分化和淋巴结转移密切相关(P<0.05);PCAF 在食管癌组织中的阳性表达为28.89%,显著低于在正常食管组织中的92.22%(P<0.05);食管癌组织中Survivin蛋白乙酰化水平与PCAF水平无相关性(P>0.05)。【结论】Survivin蛋白在食管癌组织内发生了乙酰化,且食管癌的发生、发展可能与Survivin蛋白的乙酰化程度密切相关;Survivin蛋白的乙酰化与乙酰基转移酶PCAF无相关性。【Objective】To investigate the relationship between the acetylation level of Survivin protein and the expression of P300/CBP-related factor (PCAF) and its clinicopathological features in esophageal cancer.【Methods】The acetylation of Survivin protein in 90 cases of esophageal carcinoma and adjacent normal esophageal epithelial tissues was detected by immunoprecipitation and Western blot.The expression of PCAF in esophageal carcinoma was detected by immunohistochemical staining,and the relationship between Survivin protein acetylation and clinicopathological features and the expression of PCAF protein was analyzed.【Results】The acetylation rate of Survivin protein in esophageal cancer tissues was (72.67±1.25)%,significantly higher than that in normal esophageal tissues adjacent to cancer (11.18±2.16)%(P<0.05).The acetylation level of Survivin protein in esophageal cancer tissues was closely related to TNM stage,tissue differentiation and lymph node metastasis (P<0.05).The positive expression of Survivin was 28.89%,which was significantly lower than 92.22% in normal esophageal tissues (P<0.05).There was no correlation between the acetylation level of Survivin protein and PCAF level in esophageal cancer tissues (P>0.05).【Conclusion】Survivin protein acetylated in esophageal cancer tissue,and the occurrence and development of esophageal cancer may be closely related to the degree of acetylation of Survivin protein;Survivin protein acetylation and acetyltransferase PCAF have no correlation.
