摘 要:目的 探讨共刺激分子B7-H3和增殖细胞核抗原(PCNA)在儿童骨肉瘤组织中的表达及其与患者预后生存之间的关系。方法应用免疫组织化学法(S-P法)检测2013年1月至2015年12月河北医科大学第三医院手术切除的52例儿童骨肉瘤标本组织中B7-H3和PCNA的表达,并比较B7-H3和PCNA表达与患者临床指标之间的关系。结果52例儿童骨肉瘤患者中,B7-H3和PCNA阳性表达率分别为69.2%和67.3%。B7-H3和PCNA在骨肉瘤不同分化级别、Ennecking分期、是否复发中的表达,差异有统计学意义(P<0.05)。B7-H3和PCNA两个指标之间存在正相关性(r=0.359,P=0.009)。B7-H3、PCNA同时高表达者总体生存率低于两者同时低表达者(P=0.025)。结论B7-H3和PCNA在骨肉瘤患者组织中呈异常高表达,B7-H3和PCNA均阳性的患者复发风险增加,预后较差,联合检测B7-H3和PCNA可以用于评估骨肉瘤患者的预后。Objective To investigate the expression of costimulatory molecule B7-H3 and proliferating cell nuclear antigen (PCNA) in osteosarcoma tissue of children and its relationship with prognosis of patients.Methods The expressions of B7-H3 and PCNA in pathologic specimens of osteosarcoma and bone fibrous dysplasia tissues from 52 patients receiving resection in the Third Hospital of Hebei Medical University were evaluated by immunohistochemical assay.Results The positive expression rates of B7-H3 and PCNA in the patients were 69.2%and 67.3%, respectively.Tumor B7-H3 and PCNA expression was significantly associated with differentiation grade, Ennecking stage and metastasis status ( P < 0.05).There was a positive correlation between B7-H3 and PCNA ( r = 0.359, P = 0.009).The overall survival rate of those with high expression of B7-H3 and PCNA was lower than those with low expression of both ( P = 0.025).Conclusions Overexpression of B7-H3 and PCNA are found in osteosarcoma tissues of children.High expression of B7-H3 and PCNA predicts poor survival and increases the risk of recurrence in these patients.Therefore, the combination detection could be important in detecting the occurrence and development of osteosarcoma in children.
