视网膜母细胞瘤组织中PTGER4的表达及临床意义

摘　　要：目的探讨前列腺素E2受体4(PTGER4)在视网膜母细胞瘤(Rb)组织中的表达及临床意义。方法收集本院2012年1月至2016年12月病理确诊且临床资料完整的Rb组织67例及同期正常视网膜组织21例,采用实时定量PCR和免疫组化检测PTGER4在上述组织中的表达水平,分析PTGER4表达与Rb临床病理特征(年龄、性别、侧别、IIRC分期、病理分化和视神经浸润)的关系。结果67例Rb组织中的PTGER4 mRNA水平和阳性表达率分别为2.653±1.056和56.7%(38/67),均高于正常视网膜组织的1.027±0.346和28.6%(6/21),差异有统计学意义(P<0.05)。Rb组织中PTGER4表达与年龄、性别和侧别无关,而与IIRC分期、病理分化和视神经浸润有关,其中临床分期较晚、低未分化及视神经浸润对应组织中PTGER4 mRNA水平和阳性表达率均较高(P<0.05)。结论PTGER4在Rb组织中表达升高,且与Rb的临床分期、病理分化及视神经浸润有关,PTGER4可作为判断RB浸润及病情评估的潜在指标。Objective To investigate the expression of prostaglandin E2 receptor 4(PTGER4)in retinoblastoma(Rb)tissues and its clinical significance.Methods From January 2012 to December 2016,67 Rb tissues with complete clinical data and 21 normal retinal tissues were collected.The expression of PTGER4 in the above tissues was detected by real-time quantitative PCR and immunohistochemistry.The relationship between expression of PTGER4 and clinicopathological parameters(age,sex,side,IIRC stage,pathological differentiation and optic nerve infiltration)of Rb was analyzed.Results PTGER4 expression level and positive rate in 67 Rb tissues were 2.653±1.056 and 56.7%(38/67),higher than 1.027±0.346 and 28.6%(6/21)in normal retina tissues with significant difference(P<0.05).The expression of PTGER4 in Rb was not related to age,sex and side,but related to IIRC stage,pathological differentiation and optic nerve infiltration.The PTGER4 level and positive expression rate were higher in the corresponding tissues with advance IIRC stage,low undifferentiated and optic nerve infiltration(P<0.05).Conclusion PTGER4 expression is increased in Rb tissues,which is related to the IIRC stage,pathological differentiation and optic nerve infiltration of Rb.PTGER4 can be used as a potential index for judging Rb infiltration and evaluating the condition of Rb.
