微小RNA-4286在非小细胞肺癌组织的表达及临床意义

摘　　要：目的探讨非小细胞肺癌(NSCLC)组织中微小RNA(miRNA,miR)-4286的表达与NSCLC临床病理特征的相关性。方法采用原位杂交和实时定量聚合酶链反应(Real-time PCR)方法分别检测组织芯片中140例NSCLC组织和64例新鲜冻存组织标本中miR-4286的表达,并分析其表达水平与NSCLC临床病理特征的关系,计量资料以均值±标准差(Mean±SD)表示,采用独立样本的秩和检验,计数资料采用χ^2检验。结果组织芯片中NSCLC癌组织的miR-4286阳性表达率为62.14%(87/140),明显高于癌旁配对正常组织的4.29%(6/140,χ^2=105.634,P<0.05);癌组织中miR-4286的表达与NSCLC的肿瘤大小、淋巴结转移及TNM分期密切相关(χ^2=11.393、4.586、5.530,P<0.05),而与患者的性别、年龄、肿瘤组织类型、分化程度均无明显相关(χ^2=0.496、0.003、0.067、1.399,P>0.05)。Real-time PCR方法结果显示,NSCLC癌组织中miR-4286的表达水平为0.032(0.024~0.057),显著高于配对癌旁正常组织的0.027(0.018~0.041,Z=-3.956,P<0.05),miR-4286的高表达水平与NSCLC的TNM分期、淋巴结转移明显相关(Z=-2.006、-2.016,P<0.05)。结论NSCLC组织中miR-4286的高表达与NSCLC的疾病进展与转移潜能明显相关。Objective To investigate the relationship between the expression of microRNA(miRNA,miR)-4286 and the clinicopathological features in non-small cell lung cancer(NSCLC).Methods The expressions of miR-4286 in NSCLC tissue microarray and fresh tissue cryopreserved samples were detected by in situ hybridization and real-time quantitative polymerase chain reaction(Real-time PCR),respectively.The relationship between miR-4286 and clinicopathological features in NSCLC was analyzed.Results The positive expression rate of miR-4286(62.14%,87/140)was significantly higher than that of normal adjacent tissues(4.2%,6/140;χ^2=105.63,P<0.05).The expression rate of miR-4286 in NSCLC cancer tissues was closely related to the tumor size,lymph node metastasis and TNM stage(χ^2=11.393,4.586,5.530,P<0.05),but not with the patient’s gender,age,tumor tissue type,and cell differentiation(χ^2=0.496,0.003,0.067,1.399,P>0.05).The results from Real-time PCR showed that miR-4286 expression level was highly observed in NSCLC tissues[0.032(0.024-0.057)]when compared with that in normal adjacent tissues[0.027(0.018-0.041),Z=-3.956,P<0.05].Moreover,higher expression level of miR-4286 was correlated with TNM stage and lymph node metastasis(Z=-2.006,-2.016,P<0.05)in NSCLC.Conclusion The high expression of miR-4286 in NSCLC was associated with the disease progression and metastatic potential.
