摘  要:目的探讨micrcRNA-218在激素依赖性与激素非依赖性前列腺癌细胞株中的表达。方法运用实时荧光定量方法检测microRNA-218在激素依赖性前列腺癌LNCaP细胞株与激素非依赖性前列腺癌C4-2、PC-3及DU-145细胞株中的相对表达,验证并分析其差异性。结果MicroRNA-218在C4-2、PC-3及DU-145三种激素非依赖性前列腺癌细胞株中表达差异无统计学意义(P>0.05),但均显著高于激素依赖性前列腺癌细胞LNCaP细胞株中的表达(P<0.01)。结论MicroRNA-218参与了前列腺癌由激素依赖性向激素非依赖型的进展,有望成为预测前列腺癌进展的生物标志物。Objective To verify and analyze the expression of microRNA-218 in hormone - dependent and hormone-independent prostate cancer cell lines. Methods Specific SYBR Green quantitative real - time polymerase chain reaction ( RT-PCR) was used to detect the relative expression of microRNA-218 in hormone-dependent prostate cancer LNCaP cell line and hormone-independent prostate cancer C4-2, PC-3 and DU-145 cell lines, and the differences were verified and analyzed. Results The expression of MicroRNA-218 was not different in C4-type, PC-3 and DU-145 three hormone- independent prostate cancer cell lines(P > 0. 05), but was significantly higher than that in hormone- dependent prostate cancer cell line LNCaP ( P < 0. 05 ). Conclusions MicroRNA-218 is involved in the progression from steroid-dependent to hormone-independent prostate cancer and is expected to become a biomarker for predicting the progression of prostate cancer.
