摘　　要：目的:观察直肠癌组织中VEGF-C表达与临床病理参数的相关性及与磁共振成像功能学及表征的相关性,探讨其在直肠癌诊断及预后分析中的应用价值。方法:随机选取2017年1月至2018年1月在中国医科大学附属盛京医院经手术病理确诊为直肠癌的患者35例,采集癌组织及配对的正常直肠组织和癌旁正常直肠组织,进行qRT-PCR及Western blot检测,观察VEGF-CmRNA及蛋白在直肠癌组织及配对正常直肠组织和癌旁组织中的表达水平差异、VEGF-C蛋白表达与临床病理参数相关性分析、VEGF-C蛋白表达与磁共振成像ADC值及TIC分型的关系。结果:与正常组及癌旁组比较,癌组织组VEGF-CmRNA及蛋白表达水平较高(P<0.0001、P<0.0001)。VEGF-C蛋白表达水平与直肠癌淋巴结转移具有相关性(P<0.001),与患者年龄、性别、肿瘤直径、浸润深度、分化程度、血管或神经浸润及TNM分期无相关性(P>0.05)。VEGF-C表达水平与ADC值为负相关性(P<0.0001)。VEGF-C表达水平与TIC分型无相关性(P>0.05)。结论:VEGF-C与直肠癌的淋巴结转移及磁共振的ADC值具有相关性,可以与磁共振结合共同对直肠癌淋巴结转移及预后进行预测。Objective:To observe the correlation between VEGF-C expression and clinical pathological data and the functional morphological characteristics of MRI in rectal cancer tissues,to discuss the value in rectal cancer diagnosis and prognosis.Methods:Thirty-five patients diagnosed as rectal cancer was selected from January 2017 to January 2018 in Shengjing Hospital of China Medical University,cancer tissues and paired normal rectal tissue and adjacent normal rectal tissues were collected.The different expression of VEGF-C mRNA and protein in these tissues were detected and compared by qRT-PCR and Western blot.The correlation between VEGF-C protein expression and clinical pathological characteristics or ADC value and TIC types of MRI were observed.Results:Compared with normal group and adjacent cancer tissue group,VEGF-C mRNA and protein expression were higher( P <0.000 1,P <0.000 1).VEGF-C protein expression was correlated with lymph node metastasis of rectal cancer( P <0.001),and was not correlated with patient age,gender,tumor diameter,invasion depth,the differentiation degree,vessel or nerve invasion and TNM stage( P >0.05).VEGF-C expression was negatively correlated with ADC value ( P <0.000 1).VEGF-C expression was not correlated with TIC type( P >0.05).Conclusion:VEGF-C is correlated with rectal cancer lymph node metastasis and MRI ADC value,VEGF-C may combined with MRI to forecast the lymph node metastasis and prognosis.
