摘　　要：目的探讨p62蛋白在大肠癌中的表达水平及对大肠癌细胞生物学特性的作用。方法收集60例大肠癌患者的大肠癌组织及其相应的癌旁组织标本,采用免疫组织化学染色法检测不同组织中p62蛋白的表达情况,分析p62蛋白的表达情况与患者临床特征及预后的关系。体外培养处于对数生长期的大肠癌HT29和SW480细胞,并选择p62蛋白表达水平高的HT29细胞进行转染,将转染p62特异性干扰小RNA(siRNA)的HT29细胞作为siRNA/p62组,未进行处理的HT29细胞作为阴性对照组,转染阴性对照序列的HT29细胞作为siRNA/SCR组。采用蛋白质印迹法(Westernblot)检测转染后大肠癌HT29和SW480细胞中p62蛋白的表达情况,噻唑蓝(MTT)法和划痕实验分别检测转染后HT29细胞的增殖情况和迁移能力。结果大肠癌组织中p62蛋白的阳性表达率为66.7%,明显高于癌旁组织的36.7%(P﹤0.01)。不同远处转移情况的大肠癌患者大肠癌组织中p62蛋白的阳性表达率比较,差异有统计学意义(P﹤0.01);不同年龄、性别、肿瘤直径、分化程度和临床分期的大肠癌患者大肠癌组织中p62蛋白的阳性表达率比较,差异均无统计学意义(P﹥0.05)。p62蛋白阳性表达患者的1年累积生存率明显低于p62蛋白阴性表达的患者(P﹤0.01)。HT29细胞中p62蛋白的表达水平高于SW480细胞。转染后,siRNA/p62组HT29细胞中p62蛋白的表达水平明显低于siRNA/SCR组和阴性对照组,siRNA/p62组HT29细胞的迁移距离、光密度(OD)值均明显低于阴性对照组和siRNA/SCR组(P﹤0.01)。结论p62蛋白可能在大肠癌的发生、发展过程中发挥着重要的作用,其可能是大肠癌进展和预后不良的潜在标志物。Objective To investigate expression of p62 protein in colorectal cancer and its effect on biological characteristics of colorectal cancer cells.Method Sixty cases of colorectal cancer tissues samples and their corresponding adjacent tissues samples were collected.The p62 protein expression in colorectal cancer tissues and adjacent tissues by immunohistochemistry were detected.The relationship between the expression of p62 protein and clinical characteristics and prognosis of patients was analyzed.The colorectal cancer HT29 and SW480 cells in logarithmic phase were cultured in vitro.HT29 cells with high protein expression level of p62 protein were selected for transfection,and HT29 cells transfected with small interfering RNA(siRNA)specific was used as the siRNA/p62 group,the untreated HT29 cells used as negative control group,or transfected with the negative control sequence HT29 cells as siRNA/SCR group siRNA/SCR group.The p62 protein expression in colorectal cancer HT29 and SW480 cells transfected with siRNA by Western blot.Methylthiazolyldiphenyl-tetrazolium bromide(MTT)was used to detect the proliferation of three groups of cells,and scratch test was used to detect the migration ability of three groups of cells.Result The p62 protein positive expression rate in colorectal cancer tissue is significantly higher than that in tissue adjacent to carcinoma(66.7%vs 36.7%,P<0.01).The positive expression of p62 protein in colorectal cancer tissues of patients with different distant metastasis was significantly different(P<0.01).But there was no statistically significant difference in p62 protein positive expression in colorectal cancer tissues of patients with different age,gender,tumor diameter,differentiation degree and clinical stage(P>0.05).The 1-year cumulative survival rate of patients with positive expression of p62 protein was lower than that of patients with negative expression of p62 protein(P<0.01).The expression level of p62 protein in HT29 cells was higher than that in SW480 cells.After transfection,the e
