摘 要:目的探讨在乏氧条件下,低氧诱导因子-1α(HIF-1α)和存活蛋白(survivin)在宫颈鳞状细胞癌SiHa细胞中的表达情况及与放射敏感性的关系。方法采用Western blot法检测未乏氧(空白对照组)和乏氧后24 h(阴性对照组)时SiHa细胞中HIF-1α及survivin的表达情况。采用siRNA技术转染SiHa细胞,构建HIF-1α(HIF-1αsiRNA组)、survivin(survivin siRNA组)表达沉默细胞株。分别对4组细胞进行不同剂量的照射,通过苏木精-伊红(HE)染色观察细胞形态,并比较4组细胞的放射敏感性。结果Western blot法检测乏氧培养后SiHa细胞中HIF-1α和survivin蛋白表达均升高,其中HIF-1α在未乏氧时明显低表达,乏氧后表达明显升高。HE染色结果显示,HIF-1αsiRNA组、survivin siRNA组细胞放疗后形态发生明显改变。4组随着剂量增加,放射增敏比(SER)、集落形成率(PE)、细胞的存活分数(SF)值均下降。在放射剂量为2 Gy和4 Gy时,HIF-1αsiRNA组与survivin siRNA组的SER均高于空白对照组和阴性对照组。结论乏氧环境可以上调宫颈鳞状细胞癌SiHa细胞中HIF-1α和survivin的表达,沉默HIF-1α和survivin的表达对SiHa细胞具有放射增敏作用。Objective To investigate the relationship between the radiosensitivity and expression of hypoxia-inducible factor-1α(HIF-1α)and survivin in SiHa cells of cervical squamous cell carcinoma(SCC)under hypoxic conditions.Method Western blot was used to detect the expression of HIF-1αand survivin in SiHa cells under non-hypoxic(blank control group)or hypoxic conditions for 24 h(negative control group).SiHa cells were transfected with siRNA technology to construct HIF-1αsilencing(HIF-1αsiRNA group)and survivin silencing(survivin siRNA group)cell lines.The four groups of cells were exposed to different doses of irradiation,after which the morphology of the cells was observed by hematoxylin and eosin(HE)staining,and the radiosensitivity of these cells were compared.Result Western blot assay showed that the expression of HIF-1αand survivin protein in SiHa cells cultured under hypoxic conditions increased significantly,HIF-1αshowed low expression under normoxic conditions,while was highly expressed after hypoxia.HE staining results indicated that the morphology of cells in HIF-1αsiRNA group and survivin siRNA group demonstrated significant change after radiotherapy.As the radiation dose increased,the values of sensitivity enhanced ratio(SER),plating efficiency(PE),survival fraction(SF)in all the 4 groups decreased consistently.When irradiated at 2 Gy and 4 Gy,the SER in HIF-1αsiRNA group and survivin siRNA group were higher compared to blank control group and negative control group.Conclusion Hypoxic environment can increase the expression of HIF-1αand survivin in SiHa cells of cervical SCC,additionally,silencing the expression of HIF-1αand survivin is of radiosensitizing effect on SiHa cells.
