摘 要:目的探讨细胞因子诱导的凋亡抑制因子1(CIAPIN1)基因和Fibulin-3在膀胱尿路上皮癌中的表达及其与临床病理特征之间的关系。方法收集膀胱癌及同一患者的癌旁标本98例,以癌旁组织作为对照,分别用实时定量PCR(qRT-PCR)检测组织样本中CIAPIN1和Fibulin-3基因的mRNA变化,利用Western blot检测组织样本中CIAPIN1和Fibulin-3蛋白表达情况;分析两者在膀胱癌临床病理表达特征之间的关系及两个基因的相关性。结果CIAPIN1和Fibulin-3的mRNA水平在膀胱癌组织中的表达显著高于癌旁组织(P<0.01),CIAPIN1在膀胱癌组织和癌旁组中的表达分别为4.223±0.986和2.054±0.866(P<0.05);而Fibulin-3在膀胱癌组织和癌旁组中的表达率分别为5.284±1.586和1.981±0.937(P<0.05);Western blot结果显示,与癌旁组织相比,CIAPIN1和Fibulin-3的蛋白水平在膀胱癌组织中的表达显著上调(P<0.05)。CIAPIN1和Fibulin-3基因在膀胱癌中的表达与病理分级、临床分期和淋巴转移相关(P<0.05),两者均与患者年龄和性别无关(P>0.05)。膀胱癌组织中CIAPIN1和Fibulin-3的mRNA表达呈正相关(r=0.2577,P<0.05)。结论CIAPIN1和Fibulin-3在膀胱癌中的表达均显著上调,联合检测可能增加膀胱癌诊断的准确率。Objective To investigate the expressions of CIAPIN1 and Fibulin-3 in bladder cancer and their relationship with the clinicopathological features of bladder cancer.Methods A total of 98 bladder cancer tissues and the paracancerous tissues were collected.The mRNA and protein expressions of CIAPIN1 and Fibulin-3 were detected with real-time quantitative PCR(qRT-PCR)and Western blot respectively.The relationship between the clinicopathological features and the two genes and the correlation between the two genes themselves were analyzed.Results The mRNA expressions of CIAPIN1 and Fibulin-3 were significantly higher in bladder cancer tissues than in paracancerous tissues(P<0.01).The mRNA expression of CIAPIN1 was 4.223±0.986 in bladder cancer tissues and 2.054±0.866 in paracancerous tissues(P<0.05),while the mRNA expressions of Fibulin-3 was 5.284±1.586 in bladder cancer tissues and 1.981±0.937 in paracancerous tissues(P<0.05).Western blot showed that the protein expressions of CIAPIN1 and Fibulin-3 were significantly up-regulated in bladder cancer tissues than in paracancerous tissues(P<0.05).The expressions of CIAPIN1 and Fibulin-3 were related to the pathological grade,clinical stage and lymphatic metastasis of bladder cancer(P<0.05),but not to patients'age or sex(P>0.05).In paracancerous tissues,there was a positive correlation between CIAPIN1 and Fibulin-3 mRNA expression(r=0.257 7,P<0.05).Conclusion The expressions of CIAPIN1 and fibrin-3 are significantly up-regulated in bladder cancer,and combined detection of CIAPIN1 and fibrin-3 may increase the accurate diagnosis rate of bladder cancer.
