摘  要:目的探讨瞬时受体电位锚蛋白-1(transient receptor potential ankyrin-1,TRPA1)对浸水束缚应激(water immersion restraint stress,WIRS)诱导的大鼠回肠黏膜损伤的影响。方法雄性Wistar大鼠24只,随机分为正常对照组(NC组)、WIRS回肠黏膜损伤模型组(WC组)、HC1组和HC2组。后3组大鼠放入鼠笼内,置(19±1)℃水中,水面平剑突。HC1组和HC2组WIRS前1 h腹腔注射TRPA1特异性拮抗剂HC-030031,剂量分别为100和200 mg/kg,WIRS 6 h后取背根神经节(DRG)和回肠组织。HE染色观察回肠黏膜病理损伤情况。RT-PCR、蛋白印迹法、免疫组织化学法检测NC组和WC组大鼠DRG和回肠组织中TRPA1的表达。结果与NC组比较,WC组回肠黏膜损伤严重,评分明显增高,DRG和回肠组织中TRPA1表达明显上调(P <0.001);与WC组比较,HC1组和HC2组回肠黏膜损伤情况明显好转,评分明显降低(P <0.01),且HC2组比HC1组损伤评分更低(P <0.01)。结论应激诱导大鼠回肠黏膜损伤与TRPA1表达上调密切相关。HC-030031有效减轻应激诱导的回肠黏膜损伤,并具有剂量依赖性。Objective To explore the effects of TRPA1 on water immersion restraint stress(WIRS)-induced ileal mucosal lesion(IML)in rat model. Methods Twenty-four male Wistar rats were randomly divided into four groups. NC group is untreated. Rats were fixed and immersed up to the xiphoid process in restraint devices of water bath at(19 ± 1)℃ for inducing ileal mucosal lesions in WC,HC1 and HC2 group. Rats were intraperitoneally injected 100 or 200 mg/kg of HC-030031 respectively at 1 h before WIRS in HC1 and HC2 group. DRG and ileum were isolated 6 h after WIRS. IML was observed by HE staining. The expression of TRPA1 in DRG and ileum in NC and WC group was detected by using RT-PCR,western blot and immunohistochemistry. Results Compared with those in NC group,IML was observed and TRPA1 expression was up-regulated in DRG and ileum in WC group. Compared with that in WC group,IML in HC1 and HC2 group was significantly improved,particularly in HC2 group. Conclusion Stress-induced IML was closely related to the up-regulated expression of TRPA1. HC-030031 can strongly alleviate stress-induced IML in a dose-dependent manner.
