摘  要:目的:探究准确高效测定鳖甲水溶性蛋白含量的方法。方法:以水为溶剂超声提取鳖甲蛋白,通过考马斯亮蓝G-250染色法、BCA(Bicinchonininc Acid)法、紫外吸收(UV)法对提取液中蛋白含量进行测定,并与凯氏定氮法对比。结果:蛋白浓度在0.4~1.5 mg/mL范围内,与凯氏定氮法的测定结果间的偏差分别为考马斯亮蓝G-250染色法:33%~48%,BCA法:5%~16%,紫外吸收法:>96%。结论:BCA法测定结果较准确,快速,消耗样品少,更适于鳖甲水提液中蛋白含量的测定。Objective:To explore an accurate and effective method of the determination for water soluble protein inTrionyx sinensis Carapace.Methods:TheTrionyx sinensis Carapace was extracted by the ultrosonic method,and Coomassie brilliant blue G-250 assay,bicinchonininc acid assay,UV spectrophotometric and contras Kjeldahlt was used for quantify of water soluble protein.Results:within the concentration of 0.4-1.5 mg/mL,the results of determinate deviation were 33%-48% by Coomassie brilliant blue G-250 assay,5%-16% by bicinchonininc acid assay,and > 96% by UV absorption.Conclusion :The determination of water soluble protein content in water extract of Trionyx sinensis Carapace using bicinchonininc acid method is accurate < rapid,and less consumption of samples,so it is optimum for the determination of water soluble protein content in water extract of Trionyx sinensis Carapace.
