摘  要:目的 检测肾上腺髓质素(adrenomedullin,ADM)在不同组织和手术前后的差异化表达。方法 通过免疫组化和RT-PCR检测20例骨肉瘤组织和瘤旁组织,10例正常组织中ADM表达情况;放射免疫法检测20例骨肉瘤患者术前、术后2周及10例正常人血浆中ADM的含量。结果 ADM在人骨肉瘤组织呈强阳性表达,骨肉瘤组织中的ADM mRNA表达量明显高于瘤旁组织和正常组织( P <0.05)。骨肉瘤患者术前血浆ADM含量高于健康人血浆ADM含量( P <0.05);骨肉瘤患者术后血浆ADM含量低于术前血浆ADM含量( P <0.05)。结论 ADM在人骨肉瘤组织中呈高表达,肿瘤经手术切除后ADM表达量随之降低,ADM有可能作为基因治疗新的作用靶点。Objective To investigate the differential expression of adrenomedullin(ADM) in osteosarcoma, paratumoral tissues and normal tissues, before and after surgical operation. Methods The expression of ADM in 20 cases of osteosarcoma tissues, 20 cases of paratumoral tissues and 10 cases of normal tissues was detected by immunohistochemistry and RT-PCR. The plasma level of ADM was detected by radioimmunoassay in 20 osteosarcoma patients and 10 normal controls before operation and two weeks after operation. Results ADM was strongly positive in human osteosarcoma, and the level of ADM mRNA in osteosarcoma tissues was significantly higher than that in paratumoral tissues and normal tissues( P <0.05). The plasma ADM level of osteosarcoma patients before operation was higher than that of healthy controls( P <0.05). ADM level in osteosarcoma patients was decreased after operation( P <0.05). Conclusion ADM is highly expressed in osteosarcoma tissues, and ADM expression is decreased after tumor resection. ADM may become a new target for gene therapy.
