摘  要:目的:观察人参皂苷Rg1对心跳骤停后小鼠心肌细胞正五聚蛋白3(Pentraxin-3,PTX3)表达的影响,并探讨其可能的作用机制。方法:所有小鼠随机分成4组,分别是空白对照组、人参对照组、人参干预组和模型组。取心肌组织行常规病理切片检查小鼠心肌组织病理学改变、酶联免疫吸附试验(ELISA法)检测小鼠外周血中白细胞介素-17A(Interleukin-17A,IL-17A)、PTX3、肌钙蛋白(TnT)、肌酸激酶(CK)、TNF-α、IL-6和IL-1β浓度;逆转录DNA聚合酶链反应(RT-PCR法)测定心肌组织IL-17A和PTX3 mRNA的表达;蛋白印迹法(Western bloting)测定心肌组织中IL-17A和PTX3蛋白含量。结果:与模型组比较,人参干预组小鼠外周血中IL-17A、cTnT、CK、TNF-α、IL-6和IL-1β浓度均显著降低(均P<0.05),心肌细胞PTX3 mRNA表达和蛋白含量均显著升高(均P<0.05)。结论:人参皂苷Rg1具有对抗心跳骤停后心肌缺血再灌注损伤的作用,其机制可能与激活PTX3从而抑制了IL-17A的表达有关。Objective:To observe the effect of ginsenoside Rg1 on the expression of Pentraxin-3(PTX3) in cardiac myocytes of mice after cardiac arrest and to explore its possible mechanism. Methods:All mice were randomly divided into four groups. The myocardial tissue was taken for routine pathological examination,and concentrations of interleukin-17 A(IL-17 A),PTX3,troponin(TnT),creatine kinase(CK),TNF-a,IL-6 and IL-1 beta in peripheral blood of mice were detected by enzyme-linked immunosorbent assay(ELISA). The expression of IL-17 A and PTX3 mRNA in myocardial tissue were detected by reverse transcription DNA polymerase chain reaction(RT-PCR). And the content of IL-17 A and PTX3 in myocardial tissue were determined by Western blotting. Results:Compared with model group,the peripheral blood of mice of the concentration of IL-17 A,cTnT,CK,TNF-alpha,IL-6 and IL-1 beta in ginseng intervention group decreased significantly(all P<0.05),while the PTX3 mRNA expression and protein content in cardiomyocytes were significantly increased(all P<0.05). Conclusion:Ginsenoside Rg1 has protective effect against myocardial ischemia-reperfusion injury after cardiac arrest,and its mechanism may be related to activating PTX3 and inhibiting the expression of IL-17 A.
