摘 要:目的:探讨山奈酚对人胆囊癌细胞(SGC-996细胞)增殖的影响及其机制.方法:采用浓度为0、25、50、75、100、125、150、175μmol/L的山奈酚干预SGC-996细胞24、48、72h后,CCK-8法检测SGC-996细胞的体外增殖能力;干预48h后,流式细胞仪检测细胞凋亡情况,Westernblot法检测P-S6K1及P-S6等哺乳动物雷帕霉素靶蛋白复合物1(mTORC1)途径相关蛋白的表达.结果:干预24h时,浓度为0~175μmol/L的山奈酚对SGC-996细胞增殖的抑制作用不明显;干预48h时,浓度为75μmol/L及以上的山奈酚可明显抑制SGC-996细胞的增殖(P<0.05);干预72h时,浓度为50μmol/L的山奈酚也可显著抑制SGC-996细胞的增殖(P<0.05).浓度为100μmol/L山奈酚干预48h,可显著诱导细胞凋亡(P<0.05),显著降低P-S6K1和P-S6蛋白的表达水平(P<0.05).结论:在一定浓度范围内,山奈酚随浓度的增加和时间的延长而明显抑制SGC-996细胞的增殖,并诱导其凋亡,而对细胞增殖的抑制作用可能是通过影响mTORC1信号通路实现的.Objective:To study the effect and mechanism of kaemperol on proliferation of human gallbladder cancer cell line SGC-996. Methods:Cell Counting Kit (CCK-8) assay was used to evaluate the effect of kaempferol on proliferation of SGC-996 cells after treatment of 0,24,48 and 72 hours at 25,50,75,100,125,150 and 175 μmol/L.After 48-hour treatment,flow cytometry assay and Western blot was used to investigate the effect of kaempferol on apoptosis as well as mTORC1 pathway related P-S6K1 and P-S6 protein expression in SGC-996 cells,respectively. Results:Kaempferol had no significant inhibition on proliferation of SGC-996 cells in 0-175 μmol/L range after 24 h treatment.It significantly inhibited cell proliferation at concentration of 75 μmol/L and above after 48 h treatment while at 50 μmol/L and above after 72 h treatment in SGC-996 cells ( P <0.05).Karmpferol of 100 μmol/L induced apoptosis and decreased expression level of P-S6K1 and P-S6 protein significantly in SGC-996 cells after 48 h treatment ( P <0.05). Conclusion:Kaempferol inhibited the proliferation of gallbladder cancer cell line SGC-996 in a dose and time dependent manner within a certain concentration range and induced apoptosis.The inhibition of cell proliferation may be achieved by affecting the mTORC1 signaling pathway.
