摘　　要：目的探讨泛素结合酶E2T(UBE2T)基因表达对鼻咽癌细胞增殖、迁移及侵袭能力的影响。方法选取鼻咽癌细胞系5-8F细胞,随机分为对照组、空白转染组和si-UBE2T组,其中空白转染组细胞转染空白载体,si-UBE2T组细胞转染UBE2T特异性siRNA载体,采用MTT法检测各组细胞增殖情况,采用Transwell迁移侵袭试验检测各组细胞迁移和侵袭,Western blotting法检测各组细胞UBE2T蛋白表达。结果 si-UBE2T组UBE2T蛋白相对表达量为(0. 354±0. 051),明显低于对照组和空白转染组(P <0. 05);si-UBE2T组培养第1 d、3 d和6 d吸光度值均低于对照组和空白转染组(P <0. 05);对照组和空白转染组培养第1d、3d和6d吸光度值比较差异无统计学意义(P> 0. 05);si-UBE2T组细胞迁移率和穿膜细胞数分别为(0. 322±0. 040)%和(85. 12±12. 06)个,明显低于对照组和空白转染组(P <0. 05);对照组和空白转染组UBE2T蛋白相对表达量、细胞迁移率和穿膜细胞数比较差异无统计学意义(P> 0. 05)。结论 UBE2T基因表达可促进鼻咽癌5-8F细胞增殖、侵袭及转移,有望成为鼻咽癌分支靶向治疗的靶标。Objective To investigate the effect of ubiquitin conjugating enzyme E2 T( UBE2 T) gene expression on proliferation,migration and invasion of nasopharyngeal carcinoma cells. Methods Nasopharyngeal carcinoma cell line 5-8 F cells were randomly divided into the control group,blank transfection group and si-UBE2 T group,the blank transfection group cells were transfected with blank vector,si-UBE2 T group cells were transfected with UBE2 T specific siRNA vector,MTT method was used to detect the proliferation of cells in each group;The migration and invasion of the cells were detected by Transwell migration and invasion assay;The expression of UBE2 T protein in each group was detected by Western blotting method. Results The relative expression of UBE2 T protein in si-UBE2 T group was( 0. 354 ± 0. 051),which was significantly lower than that of the control group and blank transfection group( P < 0. 05);Si-UBE2 T group cultured in 1 d,3 d and 6 d absorbance values were lower than the control group and the blank transfection group( P < 0. 05) The control group and the blank transfection group cultured in 1 d,3 d and 6 d absorbance value had no significant difference( P > 0. 05);The cell migration rate and transmembrane cells number in the si-UBE2 T group were( 0. 322 ± 0. 040)% and( 85. 12 ± 12. 06),significantly lower than those of the control group and the blank transfection group( P < 0. 05);There was no significant difference between the control group and the blank transfection group in the relative expression of UBE2 T protein,cell migration rate and the number of transmembrane cells( P > 0. 05). Conclusion The expression of UBE2 T gene can promote the proliferation,invasion and metastasis of nasopharyngeal carcinoma cell line 5-8 F cell,and it is expected to be the target of targeted therapy of nasopharyngeal carcinoma.
