摘  要:目的通过口腔涂抹慢性牙周炎致病菌的方法构建一种模拟人慢性牙周炎发病的小鼠动物模型,分析小鼠免疫细胞—脾脏细胞表面分子及血清中各型细胞因子的表达情况。方法采用流式细胞术检测小鼠脾脏细胞中表面分子的表达,ELISA法检测血清中各型细胞因子的表达。采用SPSS17.0对数据进行统计学分析。结果感染四周后,慢性牙周炎组与正常对照组相比,脾脏细胞的CD11b的表达水平显著增高(P<0.01),血清中TNF-γ和IL-6表达水平升高,IFN-γ和IL-10表达水平下降。结论慢性牙周炎小鼠模型中的脾脏细胞表面分子升高,血清中细胞因子水平改变,可能与致病菌的免疫抑制有关。Objective To investigate the expression of surface molecules of spleen cells and Th1/Th2 cytokines in serum,and an experimental periodontitis model in mice was constructed by oral application of periodontal pathogens Methods Expression of surface molecules of spleen cells was detected by flow cytometry and the level of cytokines in serum was detected by ELISA. Statistical analysis was performed using SPSS 17.0. Results After four weeks of infection,the expression of CD11b in spleen cells was significantly higher in chronic periodontitis group than in control group ( P <0.01). The levels of TNF-α and IL-6 were increased in chronic periodontitis. Conclusion The expression of surface molecules of spleen cells is increased and the levels of cytokines in serum were altered in the experimental periodontitis model,which may be associated with the immune response to pathogenic bacteria.
