摘  要:目的:探索甘氨酰tRNA合成酶(glycyl-tRNA synthetase,GARS)和肺腺癌临床特征的关联。方法:我们首先从GEO、TCGA数据库中获取了肺腺癌的基因表达数据和临床信息;接着我们通过非参数检验分别分析了GARS在肺腺癌组织和正常肺组织之间的表达差别,并且我们通过免疫组化的方式探索了肺腺癌组织和正常组织中GARS的蛋白表达;然后我们分析了GARS表达水平和肺腺癌总生存期、无进展生存期之间的关系;最后我们分析了GARS表达量和肺腺癌人口学特点、生长大小、浸润转移、分期以及肿瘤恶性生物学标志物之间的关系。结果:我们发现GARS在肺腺癌中高表达,且高表达GARS的病人具有较差的总生存期和无进展生存期;而且GARS和肺腺癌的肿瘤大小、临床分期显著相关;最后我们发现GARS和一系列肿瘤恶性标志物呈正相关。结论:我们的研究表明GARS和肺腺癌恶性程度有关,高表达的GARS往往预示着较差的临床特征。Objective:To explore the correlation between glycyl-tRNA synthetase(GARS)and clinical features of lung adenocarcinoma.Methods:We first obtained the gene expression data and clinical information of lung adenocarcinoma from the GEO and TCGA databases.Then we analyzed the difference in GARS expression between lung adenocarcinoma and normal lung tissue by nonparametric test.And we explored the expression of GARS protein in lung adenocarcinoma and normal tissues by immunohistochemistry.We then analyzed the relationship between GARS expression and overall survival and progression-free survival of lung adenocarcinoma.Finally,we analyzed the expression of GARS and demographic characteristics,growth size,invasion and metastasis,and tumor malignant biomarkers of lung adenocarcinoma.Results:We found that GARS was highly expressed in lung adenocarcinoma.Patients with high expression of GARS had poor overall survival and progression-free survival.And GARS was significantly associated with tumor size and clinical stage of lung adenocarcinoma.Finally we found GARS showed positive correlations with a series of tumor malignant markers.Conclusion:Our research suggested that GARS was associated with the malignant progression of lung adenocarcinoma and highly expressed GARS often indicated poor clinical features,which will provide some inspiration for clinical coping strategies of lung adenocarcinoma.
