摘  要:目的探讨黄芪颗粒对铅损伤大鼠脑组织抗氧化能力及学习记忆能力的影响。方法 SD大鼠40只随机分为5组,分别为正常对照组、铅损伤模型组、铅损伤黄芪颗粒低、中、高剂量干预组,每组8只,除正常对照组外其它组自由饮用浓度为1 g/L的醋酸铅溶液造模4周,后铅损伤黄芪颗粒干预组按3 g/kg、6 g/kg、12 g/kg剂量进行灌胃,正常对照组和铅损伤模型组灌胃等容蒸馏水,1次/d,连续4周。采用Morris水迷宫实验测试大鼠学习记忆能力。水迷宫实验结束后大鼠断头取脑利用化学比色法试剂盒检测脑组织中的胆碱酯酶(CHE),紫外分光光度法测定丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、一氧化氮(NO)含量。原子吸收石墨炉法检测骨铅含量。结果与正常对照组相比,铅损伤造模组大鼠逃避潜伏期、运动距离长,滞留时间、穿越次数减少,SOD、CHE、GSH-Px活性降低,MDA、NO水平增加高、骨铅含量增高(P <0.05);与铅损伤造模组大鼠相比,黄芪颗粒低、中、高剂量组逃避潜伏期、运动距离缩短,滞留时间、穿越平台次数增加,SOD、CHE活性升高,MDA、NO水平降低,骨铅含量降低(P <0.05)。结论黄芪颗粒能够降低铅损伤大鼠的NO、MDA、骨铅含量,提高CHE、SOD的活力,提高大鼠定位航行和空间探索能力,从而提高了铅损伤大鼠的脑组织中抗氧化能力和学习记忆能力。Objective To investigate the effect of Astragalus Granule on learning and memory ability of leaddamaged rats. Methods 40 SD rats were randomly divided into 5 groups, normal control group, lead damage model group, lead damage astragalus granule particles high, medium and low dose of intervention group, with 8 in each group, in addition to the normal control group, other groups free drink the lead acetate solution concentration of 1 g/L building 4 weeks, astragalus granule particles lead after injury intervention group according to the 3 g/kg, 6 g/kg, 12 g/kg dose lavage. The normal control group and the lead injury model group were given equal volume distilled water once a day for 4 weeks. Morris water maze experiment was used to test the learning and memory ability of rats. The contents of cholinesterase (CHE), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and nitric oxide (NO) in brain tissues were measured by chemical colorimetry kit after the water maze experiment. Bone lead content was detected by atomic absorption graphite furnace. Results Compared with the normal control group,the escape incubation period of the rats in the lead injury modeling group was prolonged, the frequency of crossing the platform was reduced, the activity of SOD, CHE, GSH-Px was decreased, MDA and NO levels were increased, and bone lead content was increased (P < 0.05). Compared with the lead injury model group, the low, medium and high dose groups of astragalus granule had shorter evasive latency, shorter movement distance, and more time to stay and cross the platform, the activity of SOD and CHE was increased, MDA and NO levels were decreased, and the bone lead content was reduced (P < 0.05). Conclusion Astragalus Granule can reduce the content of NO, MDA and bone lead in lead-damaged rats, increase the activity of CHE and SOD, improve the ability of navigation and space exploration, and improve the learning and memory ability of lead-damaged rats.
