摘 要:目的观察miR-574-5p在卵巢组织中的表达及下调miR-574-5p表达对卵巢癌细胞株SKOV-3增殖与侵袭的影响。方法实时PCR检测41例卵巢癌组织及对应癌旁正常卵巢组织miR-574-5p的表达;随机将SKOV-3细胞分为空白对照组、阴性对照组和miR-574-5p抑制物组,分别用MTT方法与Transwell细胞侵袭实验检测各组SKOV3细胞的增殖与侵袭能力变化。结果卵巢癌组织中miR-574-5p相对表达量为1.29±0.18,显著高于癌旁正常卵巢组织的0.51±0.07(P<0.01);转染miR-574-5p抑制物后,SKOV3细胞的增殖能力明显下降,穿膜能力亦明显减弱(P<0.01)。结论卵巢癌组织中miR-574-5p的表达水平异常增高,抑制miR-574-5p的表达能够降低SKOV3细胞的增殖与侵袭。Objective To observe the expression of microRNA-574-5p in ovarian tissue and the effect of downregulation of microRNA-574-5p on proliferation and invasion of ovarian cancer cell line SKOV-3. Methods Real-time PCR was used to detect the expression of microRNA-574-5p in 41 ovarian cancer tissues and corresponding normal ovarian tissues adjacent to cancer. SKOV-3 cells were randomly divided into blank control group , negative control group and microRNA-574 -5p inhibitor group. The proliferation and invasion of SKOV3 cells were detected by MTT and transwell assay respectively. Results The level of microRNA-574-5p in ovarian cancer tissues (1.29±0.18) was significantly higher than that in normal ovarian tissues adjacent to cancer (0.51±0.07)(P<0.01). The proliferation and invasion of SKOV3 cells were significantly decreased after transfection with the inhibitor of microRNA - 574- 5p (P<0.01). Conclusion The expression of microRNA-574-5p in ovarian cancer tissues was abnormally increased , and the proliferation and invasion of SKOV3 cells was decreased by inhibiting the expression of microRNA-574-5p.
