摘 要:目的观察人结直肠癌细胞的放射敏感性及辐射对其高尔基磷酸化蛋白2表达的影响。方法体外培养人结直肠癌细胞株HCT116、SW480及正常结直肠细胞株FHC,检测GOLPH2基因转录及蛋白表达。选取指数生长期人结直肠癌细胞进行0、2、4、6、8、10Gy照射,计数细胞克隆形成率,制备细胞存活曲线。应用RT-PCR和Western blotting检测2种结直肠癌细胞株在不同剂量照射时GOLPH2基因mRNA转录水平及GOLPH2蛋白表达情况。应用Transwell侵袭实验观察不同照射剂量对人结直肠癌细胞株HCT116、SW480迁移能力的影响。结果随着照射剂量在一定范围内增高时,人结直肠癌细胞株HCT116、SW480克隆形成率逐渐下降,两种细胞株中GOLPH2基因mRNA转录水平及GOLPH2蛋白表达水平均逐渐下降。不同X射线照射剂量对人结直肠癌细胞株HCT116、SW480迁移有抑制作用,并且在8Gy、10Gy时对人结肠癌细胞株迁移能力抑制最明显。结论 X射线对人结直肠癌细胞株HCT116和SW480的增殖及迁移能力均有抑制作用,可降低人结直肠癌细胞株HCT116和SW480中GOLPH2基因mRNA转录水平及GOLPH2蛋白表达水平。Objective To observe the radiosensitivity of human colorectal cancer cells and the effect of irradiation on the expression of golgi phosphorylated protein 2. Methods Colorectal cancer cell lines HCT116, SW480 and normal colorectal cell line FHC were cultured in vitro. The transcription and protein expression of GOLPH2 gene were detected. The exponential growth phase cells were irradiated with 0Gy, 2Gy, 4Gy, 6Gy, 8Gy, 10Gy, and the cell cloning formation rate was counted to prepare the cell survival curve. The mRNA transcription levels of GOLPH2gene and the expression of GOLPH2 protein were detected by RT-PCR and Western blotting. Transwell invasion assay was used to observe the effect of different irradiation doses on transfer ability of colorectal cancer cell lines HCT116 and SW480. Results When the irradiation dose increased within a certain range, the cloning formation rate of human colorectal cancer cell lines HCT116 and SW480 decreased gradually, and the mRNA transcription level of GOLPH2 gene and the expression level of GOLPH2 protein in the two cell lines decreased gradually. Different X-ray irradiation doses had inhibitory effects on the migration of human colorec - tai cancer cell lines HCT116 and SW480, and the most obvious inhibition on the migration ability of human colon cancer cell lines was at 8 Gy and 10 Gy. Conclusion X-rays can inhibit the proliferation and migration of human colorectal cancer cell lines HCT116 and SW480, and can reduce the mRNA transcription level and GOLPH2 protein expression level of G0LPH2 gene in human colorectal cancer cell lines HCT116 and SW480.
