三七和茶黄素对成人Ⅳ期股骨头坏死表面软骨生长的影响

摘  要:目的观察三七和茶黄素对成人Ⅳ期股骨头坏死表面软骨生长情况的影响。方法收集在全髋置换手术中获得的成人股骨头表面软骨标本,将标本打碎后分离培养软骨细胞。实验分为三七组、茶黄素组、三七+茶黄素组、对照组,对照组细胞培养不添加任何药物,三七组添加30μg/mL三七粉培养,茶黄素组添加20μg/mL茶黄素培养,三七+茶黄素组添加10μg/mL三七粉+10μg/mL茶黄素培养。分别于培养第3天、第7天、第10天在显微镜下观察软骨细胞形态与生长情况,并采用噻唑蓝(MTT)法检测各组培养48h后软骨细胞增殖情况。结果培养基中可见接种细胞为小圆形细胞,并伸长形成突起状,早期多呈多角形,符合软骨细胞形态。各药物组培养第3天、第7天、第10天显微镜下的细胞形态与对照组相比没有明显差异。软骨细胞增殖活力OD值三七组为0.34±0.02,茶黄素组为0.36±0.03,三七+茶黄素组为0.37±0.01,对照组为0.36±0.03,各组间相互比较差异均无统计学意义(P均>0.05)。结论单独应用三七和茶黄素,或者两者混合物都不能改善成人股骨头表面软骨细胞的生长活性。Objective It is to observe the effect of notoginseng and the theaflavin on the growth of cartilage in adult stage IV femoral head necrosis. Methods The specimens of cartilage in adult femoral head after total hip athroplasty were collected and broken. Then the chondrocytes were isolated and cultured. The chondrocytes were divided into four groups: notoginseng group, theaflavin group, notoginseng+theaflavin group, and control group. The control group did not add any drugs in the cell culture, and the notoginseng group was supplemented with 30 μg/mL of notoginseng powder in the cell culture, the theaflavin group was added 20 μg/mL theaflavin in the cell culture, and the notoginseng+theaflavin group was added with 10 μg/mL of notoginseng powder+10 μg/mL theaflavin in the cell culture. The morphology and growth of chondrocytes were observed under microscope on the 3 rd, 7 th and 10 th day of culture, and the proliferation of chondrocytes was detected by MTT assay after cultured for 48 hours. Results The inoculated cells were small round cells in the medium, and elongated to form protrusions, which were mostly polygonal in the early stage and conformed to the morphology of chondrocytes. The cell morphology under the microscope on the 3 rd, 7 th, and 10 th day of each drug group was not significantly different from that of the control group. The OD value of chondrocyte proliferation activity was 0.34±0.02 in the notoginseng group, 0.36±0.03 in the theaflavin group, 0.37±0.01 in the notoginseng+theaflavin group, and 0.36±0.03 in the control group, compared with each other, the difference in the OD value was not significant(P>0.05). Conclusion The application of notoginseng and theaflavins alone, or combination of the two medicine does not improve the growth activity of chondrocytes on the surface of adult femoral heads.
