摘 要:目的探讨二甲双胍联合姜黄素对雄激素非依赖性前列腺癌细胞PC3生物学行为的影响。方法实验分为对照组、二甲双胍组、姜黄素组及联合组。对照组不加任何药物,二甲双胍组采用半数抑制浓度(IC50)的二甲双胍干预,姜黄素组采用IC50浓度的姜黄素干预,联合组采用IC50浓度的二甲双胍+IC50浓度的姜黄素干预。采用MMT法检测各组PC3细胞抑制率,采用膜联蛋白V-异硫氰酸荧光素(Annexin V-FITC)法检测各组PC3细胞凋亡率,采用Transwell小室检测PC3细胞侵袭转移能力,采用Western Blot检测凋亡、侵袭转移相关蛋白表达情况。结果与二甲双胍组和姜黄素组比较,联合组对PC3细胞增殖抑制作用显著增强(P<0.05);联合组细胞凋亡率、Bax/Bcl-2值较对照组、二甲双胍组、姜黄素组显著升高(P<0.05);联合组迁移侵袭细胞数、基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)蛋白表达量较对照组、二甲双胍组、姜黄素组均显著减少或降低(P<0.05)。结论二甲双胍联合姜黄素可有效抑制雄激素非依赖性前列腺癌细胞PC3增殖、侵袭转移,促进其凋亡,其机制可能与升高Bax/Bcl-2值、抑制MMP-2和MMP-9表达有关。Objective To explore the effects of metformin combined with curcumin on the biological behaviors of androgen-independent prostate cancer cell line PC3.Methods The experiment was divided into control group,metformin group,curcumin group and combined group.Control group was not given any drugs,and metformin group was given half-inhibitory concentration(IC50)of metformin for intervention,and curcumin group was given IC50 concentration of curcumin,and combined group was given IC50 concentration of metformin+IC50 concentration of curcumin.The inhibition rate of PC3 cells in each group was detected by MMT method.The apoptosis rate of PC3 cells in each group was detected by Annexin V-FITC method.The invasion and metastasis abilities of PC3 cells were detected by Transwell chamber.The expressions of apoptosis,invasion and metastasis-related proteins were detected by Western Blot.Results Compared with metformin group and curcumin group,the inhibition effects of PC3 cell proliferation were significantly increased in combined group(P<0.05).The apoptosis rate and Bax/Bcl-2 value in combined group were significantly higher than those in control group,metformin group and curcumin group(P<0.05).The quantities of migration and invasion cells and protein expression levels of MMP-2 and MMP-9 in combined group were significantly decreased or reduced compared with those in control group,metformin group and curcumin group(P<0.05).Conclusion Metformin combined with curcumin can effectively inhibit the proliferation,invasion and metastasis and promote the apoptosis of prostate cancer cell line PC3.Its mechanism may be related to the increase of Bax/Bcl-2 value and the inhibition of MMP-2 and MMP-9 expressions.
