摘 要:目的研究中药甘草水提后药渣的化学成分及抗氧化作用,促进甘草药渣的综合利用。方法采用硅胶柱色谱法、Sephadex LH-20凝胶柱色谱法、中压柱色谱法、半制备高效液相色谱法等方法对甘草水提后的药渣进行分离纯化,通过理化常数和NMR、MS波谱等方法鉴定化合物的结构;通过DPPH自由基清除活性试验对分离得到化合物的抗氧化活性进行评价。结果从甘草药渣乙醇提取物中共分离得到10个化合物,分别为:芒柄花素(1)、半甘草异黄酮B(2)、5,7-二羟基-4′-甲氧基异黄酮-7-O-β-D-吡喃葡萄糖苷(3)、芒柄花苷(4)、Isoerythrinin A(5)、染料木素(6)、2′,4′-二羟基-4-甲氧基查尔酮(7)、4,4′-二羟基-2′-甲氧基查尔酮(8)、苜蓿素(9)、甘草素(10);化合物1~8均有较强的抗氧化活性。结论化合物3~9为首次从甘草药渣中分离得到,化合物1~8有抗氧化活性。Objective To study the chemical constituents and antioxidant activity of the residues of Glycyrrhiza uralensis Fisch.after water extraction to improve the utilization rate of the residues of Glycyrrhiza uralensis Fisch.Methods The chemical constituents were isolated and purified by repeated silica gel column chromatography,Sephadex LH-20 gel column chromatography,ODS column chromatography,high pressure flash chromatography and semi-preparative HPLC,and their structures were elucidated on the basis of physicochemical constants and spectral analysis.The antioxidant activity of the compounds was evaluated by DPPH radical scavenging activity test.Results 10 compounds were separated from ethanol extract of the residues of Glycyrrhiza uralensis Fisch.and identified as formononetin(1),Semilicoisoflavone B(2),sissotrin(3),formononetin-7-O-β-D-glucopyranoside(4),isoerythrinin A(5),genistein(6),2′,4′-dihydroxy-4-methoxylchaicone(7),4,4′-dihydroxy-2′-methoxylchaicone(8),tricin(9),liquiritigenin(10).Compounds 1-8 had strong antioxidant activity.Conclusion Compounds 3-9 was isolated from the residues of Glycyrrhiza uralensis Fisch.for the first time and Compounds 1-8 had antioxidant activity.
